Objectives: Explore the influe nce of Arid1a on cell cycle and apoptosis of gastriccance r cell lines.Methods: The BAF250a protein expression level was evaluated by Western blot in AGSand MKN28gastric cell lines. AGS and MKN28gastric cell lines were usedas the cell models. Consistant ectopic Arid1a expression was introduced intothe cells by lentiviruses expression system and the effects on cell cycle andapoptosis were evaluated. The mRNA transcription levels of Arid1a、P53、Caspase-8were examined by Real-time PCR, the apoptosis and cell cycleindex were evaluated by Flow cytometry.Results: Western blot found that the protein of Arid1a (BAF250a) in the AGS cells andMKN28gastric cancer cell lines were showed a low amount of expression;Real-time PCR found that mRNA of ARID1A genes in transfected AGS cellsand MKN28gastric cancer cell were compared with non-transfected cells,which Arid1a, p53and caspase-8were significantly increased (P <0.05);Flowcytometry found by measured cell cycle that the transfected AGS cells and MKN28gastric cancer cells in G0/G1phase percentage were significantlyhigher and G2/M、S phase percentage significantly lower than non-transfectedMKN28and AGS cell,(P <0.05). Flow cytometry found by measured apoptosisthat the percentage of apoptosis about transfected MKN28and AGS cell weresignificantly higher than the non-transfected MKN28and AGS cell,(P <0.05).Conclusion: ARID1A gene was a cell cycle inhibitor in gastric cancer cell lines, andpromoted gastric cancer cell apoptosis. It may be a tumor suppressor gene ingastric cells.
