Aim To futher explorer the role of CD151in regulating endothelial cell-cell adhesionformation and cell-cell conection stability maintainance on the basis of previous study.Method1. we constructed rAAV-CD151, rAAV-QRD, rAAV-ARSA, rAAV-anti-CD151,rAAV-GFP plasmid, andtransfected them into HUVECS.48h after transfection, we usedwestern bloting to assess the target protein CD151expression.2. We used cell aggregationassay to evaluate cell-cell adhesion.3. The cells migration and invision ability was assessedby wound closure assay and transwell asay respectively.4. The tube formation on thesurface of matrigel was used to assess angiogenesis in vitro.5. The expression of protentialinvolvement of cell signaling pathways were measured by western blot.Results The CD151, CD151-QRD, CD151-ARSA gene delivery has significantly increasedthe target protein CD151expression compared with the control group and GFP group. Bycontrast, anti-CD151gene transfection has descreased the CD151expression. The CD151high expression group showed increased angiogenesis in vitro. Consistent with the assaysrelavant to the process of angiogenesis including cell invision, migration, and cell-celladhesion. All of these above was impaired in the QRD, ARSA mutation group, andinhibited in the anti-CD151transfection group. The CD151high expression group alsoshowed increased adhesion-dependent activation of RhoA, Rac, cdc42, and increasedE-cadherin expression and FAK phosphorylation. But the QRD, ARSA mutation group hasweaken these effects. The anti-CD151gene delivery group can reverse all of the effectsabove.Conclusion The CD151-integrin complex modulates the signal molecules RhoA, Rac,cdc42activation; increases E-cadherin expression; induces FAK phosphorylation; promotesE-cadherin mediated endothelial cell-cell adhesion, and ultimately accelerates the newblood vassels formation and stabilization.
