ObjectivesGestational diabetes mellitus(GDM) is abnormal glucose tolerance in any degreewhich was found in pregnancy, is an independent type of diabetes. GDM threaten thehealthy of the pregnant women, fetal and newborns. The aim of this study was to inv-estigate the variation of the dimethylarginine dimethylaminohydrolase(DDAH)’ expr-ession and enzyme activity, discuss that the offect of DDAH to result in endothelial i-njury in the placental issue of GDM, and understand it’s influence in the clinical prog-nosis of GDM.MethodsAccording to the diagnosis standard of the American Diabetes Association, selectednormal pregnant women20cases(control group), patients with GDM20cases(experimental group). Applicat-ion of immunohistochemistry method to detect the expression of DD AH-2, enzymatic method to detect the enzyme activity of DDAH-2, Elisamethod to detect the content of ADMA in the placental issue. Compared the exp ressionand enzyme activity of DDAH-2, the content of ADMA between the control gr oup andexperimental group.Results1. Compared with the control group, the S/D, RI, PI were higher in the GDM gro-up,there were no statistically significant (P>0.05); the newborn weight and the plac-entaweight both had notable statistically significant(P<0.05).2. The pathological morphology change of the placenta In the normal pregnantwomen, the placenta villus are plentiful, almost in the sa-me size. The primary villus, secondary villus and tertiary villus have completed struct ure, tidy form, distribution,alignment. The tissue is arranged content.In the GDM, the placental villus is disorder, in the different size. The villus isdysplasia, short, arranged loose. There are some chorionic capillary hemangioma, so-mevillus fibrinoid necrosis, and few extremely expanded rough endoplasmic reticulu-m,swelling mitochondria.3. In the GDM group, the expression and the enzyme activity of DDAH-2in theplacenta is lower than that in the control group; the content of ADMA is higher than th at inthe control group. The different both have statistically significant(P<0.05).Conclutions1. GDM effect the matrix uterine arterial blood stream, make the uterine arterial vascular contracting, influence the the blood supply and nutrient transport of fetus,ultimately, meke adverse effects of fetal outcomes.2. The GDM placental tissue has significant change in the morphology compared withthe normal pregnant women. The form was changed and immature, effect mater-ialtransport between fetal and matrix.3. The expression and enzyme activity of DDAH-2was decreased in the placentatissue in the GDM, lead to weaken the hydrolysis action on ADMA. Both change result inthe increase of ADMA content and the decrease of NO. All that make the en-dothelial cellinjury heavier.4. The expression and enzyme activity of DDAH-2was decreased, to make theendothelial cell injury heavier, in the GDM’placental tissue.
