| Background:Cervical cancer is one of the three malignant tumors in female reproductive organs and ranked second. It has become a common and serious disease that threats to women’s health. In recent years, the incidence of cervical cancer and precancerous lesion has increased and become the young tendency. As a result of early cervical cancer has no specific clinical symptoms, the majority found already belongs to the late cancer. Screening to detect precancerous lesions is the key of prevention. It has been recognized that how to find cervical precancerous lesions and early cancers is the most effective measures to increase survival rate and decrease the mortality rate in cervical cancer.At present, looking for tumor markers is always the research hotspot for early diagnosis and prognosis. Id1(Inhibitor of DNA binding or inhibitor of differentiation) belongs to the family of transcription factor with double helix ring structure (helix-loop-helix, HLH). It can negative regulate the cell differentiation by regulation the gene expression, so become the factor to inhibit the cell differentiation. Recent studies showed that, Id1was over expression in a variety of malignant tumors including cervical cancer. The over expression level and cancer prognosis or malignant degree has a certain relationship in cervical cancer.Objective:Cervical disease screening was performed in8000rural women aged35-59years in Qingdao Laoshan District. The liquid-based cytology test (LCT) was used firstly. Cervical biopsy and HE dying was then used in high risk patients. The rate of cervical diseases was calculated and statasised. The expression of Idl protein was further detected, and the relationship with cervical lesions was analysised. It was expected to further find the correlation between Id1and cervical lesions, to know the diagnosis value of Id1in epithelial tumors in clinical application and to explore the possibility of Id1as a new tumor marker.Methods:Cervical disease screening was performed in8000rural women aged35-59years in Qingdao Laoshan District. The liquid-based cytology test (LCT) was used firstly, then the abnormal samples with LCT were divided into ASC-US, CINI-â…¡ (LSIL), CINâ…¡-â…¢ (HSIL). The cases were further detected for HPV DNA. Biopsy under colposcope was performed in women with LCT re-detected and HPV detection. The disease of CIN or invasive cervical carcinoma(LCC) was identified by HE dying. The expression of Idl protein was detected by immunohistochenmistry in the tissues of CIN and LCC, then the positive grade was calculated by JD801photo analysis soft. The relationship of Id1expression with the degree of cervical lesions was discussed finally.Results:Among8000subjects, totally121cases were abnormal detected by LCT. Among them,9cases were ASC-US,5cases were ASC-H,57cases were LSIL,45cases were HSIL,1case was adenocarcinoma and4cases were atypical glandular cell (AGC). The positive results of pathological biopsy were45cases from121cases with cervical biopsy. Among them,17cases were CINI,14cases were CIN II,23cases were CINâ…¢,5cases were carcinoma in situ and3cases were early invasive cervical carcinoma. The CINâ…¡ and above accounted for37.19%of the total abnormal biopsy. The prevalence was56.25per10000. In different grading of CIN samples, the expression of Idl protein was in a varying degree as:ICC>CINâ…¢>CINâ…¡>CINâ… . There was no positive Id1expression in normal cervical epithelial cells, and strongly positive expression was observed in cervical cancer.Conclusion:1. Liquid-based cytology technique combined with biopsy can provide a higher rate of diagnosis for screening cervical carcinoma. The prevalence of CIN â…¡ and above was56.25per10000in Qingdao.Laoshan district women aged from35to59.2. The detection of Idl will be helpful for the prognosis assessment of cervical disease. |
