| Objectives:Investigate the mechanisms of electro-acupuncture regulating metagenome of intestinal flora of obesity mice impacted by electro-acupuncture therapy, by contrastive analysis intestinal flora and differences of gene expression of normal group^electro-acupuncture7days groupã€electro-acupuncture14days effective markedly group and obesity mice. The research opens the door to new research field of electro-acupuncture therapy obesity.Methods:In this study, we adaptive fed fifty mice of C57for two weeks, then chose ten mice randomly as normal group, which was fed with common feed; chose forty mice as model group, which was fed with high-fat diet. Eight weeks later, we selected twenty five mice as obesity mice from model group which met standards. Then we divided the obesity mice into there groups randomly:electro-acupuncture7days groupã€electro-acupuncture14days group and spontaneous recovery group. All groups were fed with common feed at the moment. In the course of treatment, we choosed "Housanli"ã€"Sanyinjiao" and "Guanyuan" as acupuncture points which were followed by switching G6805-2electric acupuncture apparatus with1OHz,1.5V, ten minuets for each time when treating. The treatment method of spontaneous recovery group was the same as electro-acupuncture group excepted electro-acupuncture therapy. We used one-way analysis of variance by applying spss17.0to identify the level of statistical significance difference of relevant indicators. We sampled feces from obesity miceã€normal groupã€electro-acupuncture7days groupã€14days group (whose weight decreasing rate were remarkable) respectively, then we extracted metagenome of intestinal flora immediately. When the extracted DNA was qualified, the four sample were send to HuaDa gene research institute of ShenZhen for further analysis. First, DNA of four sample was break into small fragments, Hiseq2000of illumina sequenced the small fragments by applying "principle of sequencing by synthesis"; then assembled the reads to contigs by applying software; next, predicted contigs’functions to obtain ORFs by using Meta Gene Annotator software; finally, we compared ORFs with several protein database, obtained gene function and species annotation.Results:1.. After treatment of14days, there is significant weight decreasing rate difference (P=0.000) between electro-acupuncture group and spontaneous recovery group. This show that electro-acupuncture can decreased weight of obesity mice remarkable.2ã€Firmicutes phyla and proteobacteria phyla enrich largely in intestinal flora of obesity mice, but bacteroides phyla significantly decrease. Those abnormal decrease or increase flora back to normal. as the therapy time prolongs. Electro-acupuncture therapy can prompt intestinal flora abundance remarkable, also.3ã€Intestinal flora of obesity mice have lower level of substance metabolism gene. Electro-acupuncture therapy can prompt ability of substance metabolism. After electro-acupuncture therapy, those abnormal lower level of gene reach even excel normal group.Conclusion:Firmicutes phyla enrich largely in intestinal flora of obesity mice play an important role on energy harvest. Electro-acupuncture therapy can lower firmicutes and promote bacteroides of obesity mice’s intestinal flora, can restore normal content and ratio of host flora, and prompt flora abundanceã€optimize micro ecological environment to achieve results of weight loss. |
