| Vitiligo is known as a chronic, common depigmentation disorder. In recent years, the incidence rate is growing. However, the etiology and pathogenesis of vitiligo is still not completely known yet. Currently, the research of Th17cell has become a hot spot. In order to investigate the role of Th17cell in the pathogenesis of vitiligo and identify the pathogenesis of vitiligo,in this study, enzyme-linked immunosorbent assay (ELISA) technique was used to detect the levels of IL-17and IL-23in peripheral blood of vitiligo patients; SYBR Green I real-time RT-PCR technique was applied to analyze the levels of TGF-β and ROR γ t mRNA expression in peripheral blood mononuclear cells (PBMC) of vitiligo patients.Objective:To determine the cytokines associated with Th17cell in the peripheral blood of vitiligo and to investigate its role in the pathogenesis of vitiligo, provide more theoretical basis in the treatment of vitiligo.Methods:According tuo the selected standard, we collected30patients of vitiligo, with15patients in active and15paitients in satble. We also collceted15healthy controls. The levels of IL-17and IL-23in serum was detected by ELISA. The levels of mRNA expression of TGF-β and ROR Y t in peripheral blood mononuclear cell (PBMC) were detected by SYBR Green I real-time RT-PCR, then analyzed the correlation with disease activity.Results:The levels of IL-17and IL-23in active and stable vitiligo were significantly higher than that of healthy control group The level in active was hinger than the level in satble (P<0.001).. The levels of IL-17and IL-23were positively correlated with the disease activity. The levels of TGF-PmRNA in active and stable vitiligo were significantly lower than that of healthy control group. The level in active was lower the the level in stable(P<0.001).The levels of RORγt mRNA in active and stable vitiligo were significantly higher than that of healthy control group. The level in active was hinger than the level in satble (P<0.001).Conclusion:The levels of IL-17, IL-23and ROR Y are significantly elevated in paitients of vitiligo. The results suggest that IL-17, IL-23and ROR γ t may play a role in the pathogenesis of vitiligo. The levels of IL-17and IL-23are positively correlated with disease acitbity and the area of lesion. |
