The Effects Of Aerobic Exercise Combind With KGM On TORCs/CREB/PGC-1A In The Form Of Hepatic Insulin Resistance Induced By High-Fat And High-carbohydrate Diet In SD Rats

Objective:To explore the intervention and interaction of aerobic exercise and KGM on the form of hepatic insulin resistance induced by high-fat and high-carbohydrate diet in rats, and discuss mechanism of the form of hepatic insulin resistance and the intervention by aerobic exercise and KGM.Method:The study objects were506-week-old male SD rats, weight180-220g. One week of adaptive feeding, then the50rats were randomly divided into5groups, those were, control group (C), high-fat and high-carbohydrate group (HF), high-fat and high-carbohydrate with KGM group (HK), high-fat and high-carbohydrate with exercise training group (HE), high-fat and high-carbohydrate with exercise and KGM group (HEK). The C group were fed with ordinary diet, while the other4groups were all fed high-fat and high-carbohydrate diet. At the same time, the HF group didn’t take exercise training, the HK group added KGM, the HE group took exercise training, and the HEK group added KGM and took exercise training; Exercise training were used60minutes’unloaded swimming for11weeks,6times per week; the adding dosage of KGM was50mg per kilogram weight,1time per day.11weeks later, drew materials on next morning of the last exercise training and keeping fasting. FPG、FINS、PGC-1a、CREB、TORCs、PEPCK and G-6-Pase in each group of rats were examined.Results:①After11weeks of high-fat and high-carbohydrate diet, compared with C group, the PEPCK and G-6-Pase contents increased significantly in HF group rats liver, meanwhile, FINS contents and HOMA-IR increased significantly (P<0.01)②Through the double factor variance analysis, aerobic exercise could significantly lower hepatic PEPCK (P<0.05) and G-6-Pase (P<0.01) contents, moreover significantly lower FPG (P<0.05)、FINS (P<0.01) contents and HOMA-IR (P<0.01). KGM could significantly lower hepatic PEPCK(P<0.01)and G-6-Pase(P<0.01)contents, and significantly lower FPG(P<0.05)、 FINS (P<0.01) contents and HOMA-IR (P<0.01). Aerobic exercise combined with KGM had significant interaction for decreasing hepatic PEPCK (P<0.01) and G-6-Pase (P<0.05) contents in rats with high-fat and high-carbohydrate diet; Although aerobic exercise combined with KGM could make FPG level in rats with high-fat and high-carbohydrate diet to be further decreased (P>0.05), but no significant difference, while that had significant interaction for FINS (P<0.01) and HOMA-IR (P<0.05), could make FINS and HOMA-IR to be further decreased.③After11weeks of high-fat and high-carbohydrate diet, compared with C group, the hepatic PGC-1a (P<0.01), CREB (P<0.05) contents increased significantly, TORCs had no significance in HF group rats; Through the double factor variance analysis, aerobic exercise could significantly decreased hepatic PGC-1a (P<0.01) contents, CREB(P<0.05) contents, while TORCs had no significance (P>0.05) in rats with high-fat and high-carbohydrate diet, but had the trend of decreaseing. KGM could significantly decrease in rats with high-fat and high-carbohydrate diet of PGC-1a and CREB (P<0.01) contents, decrease TORCs (P<0.05) content. Aerobic exercise combined with KGM had significant interaction for hapatic of PGC-1a (P<0.01), while CREB and TORCs had no significant interaction (P>0.05), but had the trend of decreasing.Conclusion:①11weeks of high-fat and high-carbohydrate diet could significantly increase hepatic PEPCK、G-6-Pase contents and HOMA-IR in rats, which shows11weeks of high-fat and high-carbohydrate diet could induce the form of hepatic insulin resistance in rats.②Aerobic exercise or KGM could significantly prevent the form of hepatic insulin resistance iiduced by high-fat and high-carbohydrate diet in rats, leading to the reduction in PEPCK^G-6-Pase contents of liver and HOMA-IR in rats with high-fat and high-carbohydrate diet. Moreover, Aerobic exercise combined with KGM could prevent the form of hepatic insulin resistance induced by high-fat and high-carbohydrate diet in rats, and had significant promotion to a certain exent. ③11weeks of aerobic exercise or KGM probably improve hepatic TORCs/CREB/PGC-1a in rats with high-fat and high-carbohydrate diet, so as to intervene in the form of hepatic insulin resistance induced by high-fat and high-carbohydrate diet in rats. Meanwhile, aerobic exercise combined with KGM had significant interaction for improving hepatic TORCs/CREB/PGC-1a in rats with the high-fat and high-carbohydrate diet.
