| Objective To affective heme oxygenase-1(HO-1) in hydrogen peroxide induced apoptosis and expression of Aquaporin-1(AQP-1) in primary alveolar epithelial type Ⅱ (AEC-Ⅱ) of rat.Methods Healthy male Sprague Dawley(SD) rats were subjected to anesthesia, tracheal intubation and open-chest to take out lung, purification, the AEC-II were cultured by1X106/ml in cell culture plate.Primary AEC-Ⅱ collected from healthy SD rats were cultured for24hours, then divided into four groups:①group-normal (treated with saline);②group-damage(treated with H2O20.2mmol/L);③group-control(treated with HO-10.5u mol/L)④group-protection (treated with HO-10.2μmol/L and H2O20.5mmol/L).Each groups were cultured for12hours after treated. The apoptosis rate were measured by Flow cytometry before treated and after1H,3H,6H,12H. The expression of AQP-1were measured by Western blotting in the same time.Result The expression of AQP-1in group-damage declined than other groups in1H,3H,6H and12H(p<0.05),and group-control was the highest in the same time(p<0.05).There was an upward trend and peaked at6H in group-control and group-protection. The expression of AQP-1of group-control and group-protect higher than group-normal and group-damage in1H,3H,6H and12H(p<0.05).Significant increase of apoptosis rate in group-damage(p<0.05). Apoptosis rate of the group-protection was not only lower than group-damage(p<0.05),but also higher than group-control and group-protection(p<0.05). No significant difference between group-control and group-protect in apoptotis rate (p>0.05)Conclution The apoptosis rate increased by the decrease of the expression of AQP-1in hydrogen peroxide primary AEC-Ⅱ of rat.HO-1could increase the expression of AQP-1and decrease the apoptotis rate in hydrogen peroxide primary AEC-Ⅱ of rat.Regulate the expression of AQP-1maybe one of the HO-1anti-apoptotic mechanism. |
