Relationship Of Gene Polymorphisms Of FABP2and Environmental Factors With NTDs

Objective:The purpose was to find out some suspectable risk factors of neural tube defects(NTDs)in Guizhou Province. Meanwhile, it was explored that the correlation between Adiponectin genel (APM1), fatty acid-binding protein2(FABP2) and uncoupling protein2(UCP2)gene in mother and susceptibility to NTDs in offspring. The interactions among these risk factors were analyzed and the functions of these factors were detected in offspring NTDs.Methods:1:1matched case-control study was used, Objects were chose from17hospitals in8regions with high incidence of NTDs in Guizhou Province. All the objects were divided into two groups:63deliveries with NTDs as a case group and63deliveries with normal newborns as a control group. All the objects were investigated in uniform questionnaires about suspectable risk factors for NTDs and were genotyped for APM145TG,FABP A54T and UCP2A55V polymorphism by(polymerase chain reaction restriction fragment length polymorphism analysis, PCR-RFLP)method. All the data were analyzed by SPSS16.0statistical software. The risk factors of NTDs were analyzed by the conditional Logistic regression.Results:Comparing APM1gene frequency between the two groups, it was showed that only three genotypes were found in APM1SNPT45G. The frequencies of TT, TG and GG were respectively39.7%,42.9%and17.5%in case group,35.6%,31.7%and12.8%in control group(χ2=3.18, P>0.05). Allele frequencies of T and G were respectively61.1%and38.9%in casegroup,61.4%and38.6%in control group(χ2=3.00, P>0.05). The discrepancy was remarkable.Comparing FABP2gene frequency between the two groups, it was showed that only three genotypes were found in FABP2Ala54Thr. The frequencies of AA, AT and TT were respectively33.3%,50.8%and15.9%in case group,55.6%,39.7%and4.8%in control group(χ2=8.13, P<0.05). Allele frequencies of A and T were respectively58.7%and 41.3%in casegroup,75.4%and24.6%in control group(χ2=7.92, P<0.05). The difference was remarkable between two groups.Comparing UCP2gene frequency between the two groups, it was showed that only three genotypes were found in UCP2A55V. The fiequencies of AA, AV and VV were respectively49.2%,44.4%and12.8%in case group,50.8%,38.1%and69.8%in control group(χ2=0.79, P>0.05). Allele frequencies of T and G were respectively65.1%and34.9%in casegroup,69.8%and30.2%in control group(χ2=0.65, P>0.05). The discrepancy was remarkable.By univariate conditional Logistic regression analysis, it was indicated that5factors were correlated with NTDs(P<0.05)in the level a=0.05. By multi-variables conditional Logistic regression analysis, it was showed that there were four factors were correlated with NTDs:vomiting of pregnancy (OR=3.560), taking germinated potato frequently (OR=3.364), passive smoking(OR=5.803), and FABP Ala54Thr mutation(OR=2.412).Analyzing interaction between factors, There was synergistic action the polymorphism of FABP Ala54Thr and vomiting of pregnancy (attributable proportions of interaction (API)=0.36, the synergy index S,(s)=1.80). And there was synergistic action between the polymorphism of FABP Ala54Thr and germinated potatoes(API=0.67, S=3.57).Conclusion:It is suggested that not APM1SNP T45G, UCP2A55V but FABP Ala54Thr polymorphism in mother is one independent genetic risk factors of offspring NTDs. Vomiting of pregnancy, taking germinated potato frequently and passive smoking in early pregnancy are the important nutritious, environmental risk factors in offspring NTDs.
