Objective:To observe the effects of adipose-derived stem cells(ADSCs) transplantation into rats infarcted myocardium at different times on cardiac function,and to explore the role of ADSCs transplantation after myocardial infarction(MI).Methods:ADSCs were isolated from the fatty tissue of homogenous Sprague-Dawly(SD) rats and cultured until fouth generation. The surface molecular markers(CD30,CD90,CD34,CD45,Sca-1,KDR) of fouth-generation ADSCs were analysed in flow cytometer(FCM).30female SD rats were ligated left anterior descending coronary artery to induce MI,then separated randomly into3groups, cells transplantation group at1week(ADSCs-1W,N=10),cells transplantation at2week(ADSCs-2W,N=10), untreated group(control,N=10). The density of ADSCs was5×106/ml. ADSCs were injected into the infracted heart of rats through epicardium. Echocardiography and Hemodynamics were measured8weeks after MI,consisted of the left ventricular end-diastolic pressure(LVEDP mmHg), the max ratio of the left ventricular pressure decay(-dp/dt max), the left ventricular ejection fraction(LVEF%),the short-axis fractional shortening(FS%), the left ventricular end-diastolic inner diameter(LVEDD cm) and heart ratio(HR b/min).Results:High expression of CD13and CD90in ADSCs with FCM detection,in contrast,low expression of KDR,Sca-1and CD34and no expression of CD45Compared with the control group,LVEDP in the both cells transplantation groups (ADSCs-1W14.60±2.66mmHg, ADSCs-2W17.78±1.84mmHg) were lower than control group(28.03±3.56mmHg,P<0.01),and-dp/dt max were increased (P<0.05). Both LVEF and FS in both cells transplantation groups were higher than control group (P<0.01). LVEDP in the ADSCs-1W group was lower than ADSCs-2W group (P<0.05).Conclusion:1.Cardiac function of myocardial infracted rats is improved or recovered partly by ADSCs transplantation.2.Improvement of cardiac diastolic function of SD rats may be better to transplant ADSCs at1week after MI.
