The Expression And Clinical Significance Of PAF、PAF-AH And PAF-R In Women With Unexplained Infertility At Implantation Window Stage

Objective:To investigate the expression and clinical significance of platelet activating factor (PAF)、platelet activating factor-acetylhyarolase (PAF-AH)and PAF-receptor (PAF-R) in women with unexplained infertility(UI) during the period of the implantation window.Methods:In this study,10women in early pregnancy(pregnancy40+days, the early pregnancy group) and20unexplained infertile women(the unexplained infertility group),10normal women(the normal group) were divided into three groups. During the period of implantation,①Elisa assay were applied to analyze the levels of PAF and the activity of PAF-AH.②Expression of PAF-R on peripheral blood monocytes (PBMCs)was examined by cytoflowmetry.③PAF-R in endometrum and decidual tissues were examined by immunohistochemistry methods.Results:①During the period of implantation, the concentration of PAF in serum(7.49±0.90)ng/ml, the activity of PAF-AH (15.37±2.11)ng/ml, the expression of PAF-R on PBMCs (40.79±15.54)%and in endometrium (0.27±0.04) in the group of the unexplained infertile women were significantly lower than those in the other two groups (the normal group [(16.84±3.50)ng/ml、(8.16±2.26)ng/ml、(75.07±8.41)%、0.36±0.05], the early pregnancy group [(33.19±4.71)ng/ml、(3.74±0.59)ng/ml、(88.07±4.08)%、0.45±0.06](P <0.01, respectively).②PAF-R on PBMCs was positively correlated with that on the endometrium in all these three groups.Conclusion:①The concentration of plasma PAF decreasing significantly, while the activity of PAF-AH increasing significantly, the expression of PAF-R on PBMCs and in endometrium reducing markedly in the group of unexplained infertility in window of the implantation, all these may be one of the mechanisms of causing unexplained infertility by decreasing the endometrial receptivity. It also indicated that the changes of PAF PAF-AH and the expression of PAF-R on PBMCs and endometrium may play a certain role in the pathogenisis of the unexplained infertility.②The expression of PAF-R on PBMCs can indirectly reflect the expression of PAF-R in the endometrium.
