ObjectiveTo study the effect of APS on hypertrophic myocardium caused byabdominal aorta constriction in rats,and to explore its molecular mechanism.MethodsTaking75rats who were suffered abdominal aorta constriction.1weekslater, these rats would be divided into4groups: myocardial hypertrophy(AAC),different dose APS treatment200,400,800mg kg-1,after accepting therapy for11weeks. Then the hemodynamics,such as left ventricular systolicpressure(LVSP),of all animals was detected with the BL-420system under thesodium.Take the tissue of heart to detect calculated heart mass index (HMI)and left ventricular mass index (LVMI) and pathological section by HE stain.The change of microtubules of myocardial cytoskeleton were detect byfluorescent immuno-assay. The contents of ATP,ADP and AMP in rat weredetected by high performance liquid chromatography (HPLC).The lactic acid(LAC)and free fatty acids (FFA) were measured. The mRNA expression ofheart atrial natriuretic peptide (ANP) was observed through RT-RCR. Isolatedcardiac cells and detect the mitochondrial membrane potential.ResultsThe heart function,ratio of ventricle weight to heart weight and ratio ofheart weight to body weight of AAC group were higher significantly than that ofSham group. Results of optical microscope: myocyte got hypertrophy, and cells arranged in bad order; ANP mRNA expression,the content of LAC andFFA was largely increased in AAC group(P<0.01);The microtubules turnedrough with no clear structure; the content of ATP,ADP and AMP was largelydecreased in AAC group(P<0.01);Compared with AAC group, the heartfunction、ratio of ventricle weight to heart weight and ratio of heart weight tobody weight of APS group reduced significantly(P<0.05or P<0.01).Microtubules changed slightly in the APS groups compared with Sham group.Results of optical microscope:the level of myocyte hypertrophy was muchlower in Pue group, and cells arranged in good order; ANP mRNA expression,the content of LAC and FFA of APS group reduced significantly than that ofAAC group(P<0.05or P<0.01); the content of ATP,ADP and AMP was largelyincreased in APS group(P<0.05or P<0.01);The expression of mitochondrialmembrane potential was effectively increased by APS(P<0.05or P<0.01).Conclusion1. APS can inhibit the progress of cardiac hypertrophy in rats.2. APS could improve the heart function fall,myocyte hypertrophy induced byabdominal aorta constriction in rats.APS has adjustment effect to relatedcytokines in myocardial hypertrophy induced by abdominal aortaconstriction.3. APS can inhibit the progress of cardiac hypertrophy,the mechanism maybe related to energy metabolism and the protection of myocardialmitochondria.
