ObjectiveTo investigate osteogenic induction of new born SD rat calvaria osteoblastson rabbit bone marrow mesenchymal stem cells(BMSCs), then,both of the cellswere co-cultured with transwell chamber.MethodBMSCs were obtained from rabbits with whole marrow method andCalvaria osteoblast were gained from born three days SD rats.Using flowcytometric analysis tested the exepression of its identification,for exampleCD44,CD34,CD45,CD90.SD rat osteoblasts and rabbit mesenchymal stem cellswere co-cultured with transellcell cchamber.Then morphological changes ofBMSCs were observed by microscope.The experimental group was vaccinatedosteoblast in upper chamber,The control group is not vaccinated cells,The twogroups were inoculated BMSCs in lower chamber.ALP activity were assessedby ELISA Reader. Genes expression of Bone Morphogenetic Protein-2(BMP-2),osteocalcin, and collagen I were investigated by RT-PCR.Result1.The morphologic changes of BMSCs presented the feature of osteoblastlike cells while control group expressed little change.2.The BMSCs expression for the ALP activity0.365±0.016, compared with0.17±0.012,ALP activity of the first group was higher than control one.3.In the first group we observed the genes expression of Bone Morphogenetic Protein-2(BMP-2), osteocalcin, and collagen I. But in the controlgroup there is no genes expression of these above materials.ConclusionCalvaria osteoblasts derived from SD rats can induce the BMSCs derivedfrom rabbits into osteoblasts successfully.BMSCs expressed the specificity ofosteoblast surface marker.
