The extraction conditions of protein from Lotus seed was optimized by DEAE-cellulose52. Further more, the nutrient value of the isolated protein was evaluated and its antioxidant effects was studied by the experiment of D-galactose Induced-Aging Mice. the results are as follows:1. the lotus seed total protein extract in the Tris-HCl buffer, and the response surface method was employed in optimizing the extraction conditions, when the liquid ratio was7.2:1, the extraction time was2.4h, and the extraction temperature was11.1℃, the total protein extraction rate was reached81.35%, and the relative error of the regression model is less than0.5%.2. Purified the total protein from lotus seed by DEAE-cellulose52, and collected much more pronounce peak eluting with different concentrations (0.2,0.4,0.6,0.8mol/L) of Tris-NaCl gradient elution, the concentration of isolated protein was83.75%. The kind of amino acids in isolated protein was16, and the proportion of essential amino acids was57%. The amino acid score was48.2and chemical score was38.4, which closed to the amino acid pattern of the WHO/FAO. The biological value was about47, which indicated the nutritional value was comprehensive.3. Using vivo experiments to explore the lotus seed isolated protein anti-aging activity. Establish the D-galactose induced aging animal models, divided into normal group, model group, high dose group, low dose group and positive control group, by measuring antioxidant indices(including malondialdehyde(MDA), catalase(CAT), superoxide dismutase(SOD), glutathione peroxidase(GSH-Px))in the blood, liver, heart and kidney and hydroxyproline content in the skin, study the anti-anging activity of the lotus seed isolated protein, the result shows, high doses of the lotus seed isolated protein(500mg/kg/d) can significantly reduce blood and viscera organization MDA content(P<0.05), but can significantly with sex increase the CAT, GSH-Px, and SOD enzyme activity and hydroxyproline content in the skin, low dose of the lotus seed isolated protein(100mg/kg/d) has not significantly effect(P>0.05) but has a certain extent effect compared with model group, the positive control group(300mg/kg/d) is almost between both reflecting the theoretical dose relationship. And the lotus seed isolated protein for the power size of the antioxidant enzymes is: CAT>SOD>GSH-Px. Cells observed in paraffin sections from the microscopic point of view clear analysis between the different groups, further confirmed the accuracy of the indicators measured by the front. Because the research report of lotus seed protein anti-aging action is very seldom, so the research results of this experiment provide a scientific basis for further development, utilization and investigation of lotus seed.
