Study on blood indicators with the XMT its various extracts of migraine model (nitroglycerin (NTG) type, induced by reserpine low blood indicators of serotonin (5-HT) type);filter total phenolic acids, and verify the effective parts with Nitroglycerin model experimentsMethods:Mixed rhizoma gastrodiae and ligusticium wallichii (4:1) by decoction collect the volatile oil, and then use different solvents to gradient extraction of XMT water decoction, get XMT various extracts;Rats as the research object, based on the NTG mode in plasma serotonin (5-HT), nitric oxide (NO), endothelin (ET), calcitonin gene related peptide (CGRP), beta endorphin (β-EP) as observation indexes, and low blood level5-HT in mice as the research object, in5-HT and5hydroxy indole acetic acid (5-HIAA) as observation index, select the efficacy of the active ingredient of treatment with migraine in Xiongma Decoction; Using ethanol refluxing extraction,extraction of XMT effects in the active site and effective active ingredient of total phenolic acids, type in the NTG rats as the research object,5-HT and NO in plasma as observation index, discuss the influence on model of rats.Results:(1) XMT medicinal materials mixed powder4560g, obtain volatile oil4ml;The materials904g obtain238g water extraction;The materials786g obtain95.4g of water extraction by alcohol sedimentation; The materials2870g obtain petroleum ether, ethyl acetate, n-butanol and water extract rest after solvent extraction are0.9g,8.7g,36.5g and613.0g.(2)NO and CGRP,5-HT, ET content of model group increased significantly compared with blank group,β-EP content significantly decreased;NO,CGRP,5-HT and ET in rats plasma of group A,E, H were decreased compared with model group (P<0.05);CGRP,5-HT of group B are decreased (P<0.05); NO, CGRP and5-HT of group F are decreased (P<0.05)(3)5-HT in rats plasma of model group lower than normal group,5-HT in the group A, B, E,F, H are increased(P<0.05);5-HIAA in rats plasma of model group lower than normal group,5-HIAA in the group A, E, H are increased(P<0.05, P<0.01).(4) through macroporous resin, the total phenolic acids in XMT accounted for40.3%of the total elution.(5) NO content group of Total phenolic acids and alcohol extract is lower than the model group(P<0.05, P<0.01).Conclusion:The ethyl acetate extract and n-butanol extract having a migraine treatment effect, can be identified as medicinal active site of Xionsma decoction. The total phenolic acid as the goal,separation and extraction of total phenolic acids in XMT accounted for40.3%of the total elution, total phenolic acid compared with model group can decrease the content of NO in rat plasma.Preliminary proved it has certain effect on the treatment of migraine.
