| Objective:This study was to observe protective effects of Liu Wei Di Huang Wan on the Wistar rats which had got non-alcoholic fatty liver disease(NAFLD) induced by a high-fat diet, and to evaluate its regulatory mechanism of non-alcoholic fatty liver disease (NAFLD) rats.Methods:Forty Wistar rats were divided into four groups at random:the normal control group (A). model control group (B). simvastatin treated group (C) and Liu Wei Di Huang Wan treated group (D). Each group had ten rats. Except that rats of he normal control group were fed with normal diet, others were high-diet to induce hyperlipidemia for8weeks. Then besides that the group B.C,and D were fed with hyperlipidemia,the rats of group C were orally given simvastatin at dose of1mg·kg-1·d-1and the rats of group D were orally given Liu Wei Di Huang Wan at dose of1.2g·kg-1·d-1for4weeks. On the last day, diets were removed from all cages14h before the rats were sacrificed and blood samples were collected and centrifuged at3000r/min for30min to get serum. The levels of serum aspartate amiotransferase (AST), alanine aminotransferase(ALT), adiponectin, low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C).insulin, albumin (ALB),glucose and total protein (TP) were determined by an automatic chemical analyzer according to the manufacturers instructions. Besides, the levels of total cholesterol (TC), triglycerides (TG), superoxide dismutase (SOD), maleic aialdehyde (MDA) in liver tissues were determined also. The protein expressions of AdipoR1and AdipoR2were detected by western blot analysis. The RNA expressions were detected by RT-PCR analysis.Results:LiuWeiDiHuangWan could effectively reduce the levels of AST, LDL-C, Glu, Ins, TG, TC and MDA.It could effectively increase the level of SOD. The histopathological analysis showed that LiuWeiDiHuangWan could significantly alleviate symptoms in the hepatopathy rats such as hepatic steatosis, lobular inflammatory cells infiltration and necrosis. It also showed that the lipid droplets in liver cells become smaller and less.Compared with the model group.the protein as well as RNA expressions of AdipoR2were obviously increased in LiuWeiDiHuangWan group as well as RNA (all P<0.05).Conclusions:LiuWeiDiHuangWan has positively therapeutical effects on rats of experimental NAFLD model. Oral administration of LiuWeiDiHuangWan to hyperlipidemia rats were highly effective in the regulating lipid metabolism, decre(?)sing lipid peroxidation up-regulating AdipoR2expressions in hepatocytes. |
