Study On Material Foundation Of Anti-rheumatoid Arthritis Efficacy And Mechanism Of Bauhinia Championii (Benth.) Benth

Objective:In present study, the effective part of a commonly used she medicine Bauhinia championii (Benth.) Benth was selected as the research object, then we investigated its anti-rheumatoid arthritic (RA) effect, and systematacially separated the chemical component to illustrate the anti-RA material basis of Bauhinia championii(Benth.) Benth, meanwhile discuss its anti-RA mechanism preliminary.Method:(1) To investigate effect of different extraction part of Bauhinia championii (Benth.) Benth on collagen-induced arthritis (CIA) rat paw swell and pathological changes of joint by building CIA model.(2) Organic solvent extraction technology, silica gel column chromatography, poly brilliant amine column chromatography and gel column chromatography technologies were employed to systematacially separated the the anti-RA effective part of Bauhinia championii (Benth.) Benth, furthermore UV, MS,1H-NMR and13C-NMR etc. modern spectrum technologies were also used to identify the chemical component structure.(3) To investigate effect of ethyl acetate extraction part of Bauhinia championii (Benth.) Benth on correlation factor expression in TLR4/NF-κB signaling pathway in CIA rat.Result:(1) Extraction of Bauhinia championii (Benth.) Benth could inhibite the paw swell of CIA rat, improve synovial tissues inflammatory infiltration and hyperplasia and so on inflammatory response, reduce the expression levels of some proinflammatory factors, such as TNF-α、IL-6、IL-8.(2) It was found that there were11chemical component in the effective part of Bauhinia championii (Benth.) Benth. It illustrated the anti-RA material basis of Bauhinia championii (Benth.) Benth.(3) Ethyl acetate extraction part of Bauhinia championii (Benth.) Benth could inhibite the paw swell of CIA rat, improve synovial tissues inflammatory infiltration and hyperplasia and so on inflammatory response, reduce the expression levels of some proinflammatory factors, such as TNF-α、IL-6、IL-8, and suppress the expression of correlation factor in TLR4/NF-κB signaling pathway, like TLR4, MyD88, NF-KB, TNF-α、IL-6、IL-8in synovial tissues.Conclusion:Bauhinia championii(Benth.) Benth provide good anti-RA effect, especially Ethyl acetate extraction part. It may be a potential anti-RA medicine, whose material basis mainly is flavonoids compounds and action mechanism maybe is to adjust correlation factor in TLR4/NF-κB signaling pathway.
