Cell-cycle analysis and cell apoptosis of bladder carcinoma cells with or without siSurvivin and15d-PGJ2treatment were examined by flow cytometry analysis (FACS). The levels of survivin expression in the bladder tumor cells with different treatments were checked by Western Blot. The spheroid-forming ability of the bladder tumor cells, as an indicator of cancer stem cell properties, was evaluated by spheroid formation assay.[Results]1.siSurvivin decreased Survivin expression in human bladder carcinoma T24and5637cells.2.15d-PGJ2significantly inhibited the proliferation of T24and5637cells.(P<0.05)3.15d-PGJ2substantially inhibited colony forming-abilities of T24and5637cells.4.15d-PGJ2treatment significantly resulted in a G2/M block in cell cycle of T24and5637cells (P<0.05)5.15d-PGJ2treatment reduced the level of Survivin protein in T24and5637cells.6.siSurvivin induced apoptosis of T24and5637cells, and15d-PGJ2could significantly enhance siSurvivin-mediated effect.(P<0.05)7.siSurvivin inhibited spheroid formation of T24and5637cells, which could be further enhanced by15d-PGJ2.(P<0.05)[Conclusions]1. siSurvivin and15d-PGJ2both can induce cell apoptosis and decrease the levels of Survivin protein of T24and5637cells.15d-PGJ2can inhibit the cell proliferation and blocks the cell-cycle progression at G2/M in T24and5637cells.2.
