Sensitized fluorescence of chlorogenic acid (CA) and ferulic acid (FA) and itsapplication in analysis of Chinese herbal medicine were studied. Fluorometric method and athin-layer sensitized fluorescence scanning method for quantitative determination of CA inChinese herbal medicine, such as Flos Lonicerae Japonicae (FLJ, Jinyinhua in Chinese) etc,were established. A new method for qualitative analysis of FA in some medicinal crops wasproposed.Three parts were included in this thesis:1. The hydrolyzation process of CA was studied. The hydrolysis product can producefluorescence which is the same as that of caffeic acid. Addition of borax and cetyl trimethylammonium bromide (CTAB) into CA solution can produce sensitized fluorescence. Thesuitable condition for the production of sensitized fluorescence was investigated, and themethod was applied to analysis of Chinese herbal medicine which contains CA.2. A new thin-layer sensitized fluorescence scanning method was proposed for theanalysis of CA in FLJ. This method was done by firstly separate the components of FLJ viathin-layer chromatography, and secondly spray borax and CTAB solution on thin-layer plateto enable CA to produce fluorescence, and then measure fluorescence intensity of the spots byfluorescence scanning. Using the method, content of CA in FLJ was measured to be3.38%and3.48%, by a standard curve method and a standard addition method, respectively. Toconfirm the reliability of the method, a solution fluorometric method and a thin-layerabsorbance scanning method were proposed and carried out, and the results were basically thesame. On this foundation, sensitized fluorescence of some herbal medicines containing CA,such as Flos Chrysanthemi (Juhua), Folium Artemisiae Argyi (Aiye), Folium Eucommiae(Duzhongye), Herba Artemisiae Scopariae (Yinchen), Flos Inulae (Xuanfuhua) and CaulisLonicerae Japonicae (Rendongteng), were studied. Using the fluorometric method and thethin-layer sensitized fluorescence scanning method, content of CA in these herbal medicineswere measured, respectively. The results of the two methods were basically the same.3. The sensitized fluorescence FA by addition of Mg2+was studied. Fluorescence of FAwas very weak. In alkaline condition (pH>10), addition of Mg2+into FA solution canenhance the fluorescence of FA more than ten times. This technique was applied in qualitativeanalysis of FA in Chinese herbal medicine, and a simple, effective, rapid method forqualitative analysis of FA was established.
