Objective:To invetst the expression level of survivin or Bcl-2protein induced by paclitaxel and cell survival rate in human ovarian cancer cell SKOV-3(MyD88+), A2780(MyD88-), which can reveal the mechanism which curcumin sensitized paclitaxel chemotherapyMethods:(1)ELISA method was used to detect the expression level of survivin or Bcl-2in ovarian cancer cells.(2)Bradford assay for the detection of total protein.expression, and pg/mg protein showed the expression level of the two target proteins.(3) MTT method was used to detecte the ovarian cancer cell growth inhibition by drugsResults:(1) Paclitaxel and LPS can promote the expression of survivin and Bcl-2on concentration and time dependent manner in the SKOV-3cells, paclitaxel could decrease the expression of two target proteins in A2780cells, However, LPS could not.(2) Curcumin could significantly decrease the expression of two target proteins at high concentrations in the both cell lines, However, the low concentration of the drug could not.(3) Compared with the drug alone, combined with curcumin on high concentrations the expression of survivin and Bcl-2could be significantly decreased, However, the low ones combined with paclitaxel or LPS only decreased survivin protein the expression, not Bcl-2.(4) Paclitaxel and curcumin can inhibit the proliferation of SKOV-3and A2780cells and decrease the IC50, coadministration with each other could significantly strengthen the effect.Conclusions:(1) In MyD88+ovarian cancer cell, paclitaxel-TLR4/MyD88+surviving/Bcl-2signaling pathway is existed.(2) Paclitaxel or LPS could activate the way to promote the over-expression of survivin/Bcl-2, however, curcumin could suppress it in time and concentration dependent maner, decresing the expression of the two proteins, especially in MyD88+ovarian cancer cell lines.(3) Curcumin could inhibit the proliferation of ovarian cancer cells and enhance the cytotoxicity of paclitaxel on non TLR4/MyD88depend way to achieve the chemotherapy sensitivity to paclitaxel.
