Background Heart valve disease is due to inflammation, myxoid degeneration, degenerative change, congenital malformation, avascular necrosis, trauma and causes of valve structure is unusual, leading to a narrow and close the valve mouth is the result of the heart disease. At present, in China and other developing countries in order to rheumatic heart disease is the most common, rheumatic inflammatory process is to valve damage, the main pathological changes is the bicuspid thickening, contracture, fibrosis and different levels of calcification, lead to valve mouth area is reduced and closed insufficiency.Among them, the mitral valve involvement is about70%.At present people to this has been doing a lot of research, although we can to prevent the recurrence of rheumatic fever success, but it can not effectively change of heart inflammation rheumatoid disease development. Therefore, further understanding of rheumatoid valve disease occurrence, development, invasive mechanism and the pathogenesis of pathological form, early diagnosis, control the progress of the disease, the change of heart inflammation rheumatoid disease development, will be the focus of research rheumatic valvular disease. Objective To analyze expressions of bFGF and BMP2in rheumatism mitral valve calcification, to investigate relationships among bFGF and BMP2and clinicopathologic factors of chosed patients and to obtain the theoretical basis for diagnosis and treatment.Methods1.Select chest, henan province hospital of thoracic surgeons July2009to September period mitral ale replacement valve specimens removed72patients, of whom rheumatism mitral valve calcification52cases, mitral valve calcification20cases of rheumatism. The same period12cases of accidental death than heart disease mitral valve tissue samples were compared.2. Using immunohistochemistry SP method, with bFGF and BMP2protein polyclonal antibody sixty cases of rheumatism mitral valve calcification resection specimen,20cases of rheumatism calcified and12cases of normal tissue specimens of paraffin wax biopsy staining, and observation, evaluation bFGF and BMP2positive rate of protein expression.3. Analysis bFGF and BMP2protein expression and clinical pathology index of the relationship.4. SPSS17.0for statistical software was used to analyze data. According to the material type, chi-square and spearman’s correlation analysis were used in statistical processing. The significant difference was considered when the P value was less than0.05.Results1. bFGF positive product mainly in cell plasma and the extracellular matrix expression, is tan-yellow particles. Normal tissue and the mitral valve calcification organization respectively with rheumatism mitral valve calcification in mitral valve organization rheumatism bFGF expression of the positive greup (χ2=18.648,P=0.000, χ2=16.744,P=0.000). According to the degree of mitral valve calcification rheumatism calcification is different, bFGF positive expression to have significant difference, but with age, sex, Pathology classification have nothing to do.2.BMP2positively product main role in cell plasma and the extracellular martrix, the nucleus without expression,is tan-yellow.Normal tissue and the mitral valve calcification organization respectively with rheumatism mitral valve calcification in mitral valve organization rheumatism BMP2expression of the positive greup (x2=9.789,P=0.005, χ2=19.871,P=0.000). According to rheumatism mitral valve calcification organization calcification degree, BMP2positive expression to have significant differences, and with age, sex, Pathology classification, gross classification have nothing to do.3.In mitral valve calcification in rheumatism disease, bFGF and BMP2expression are positively related (r=0.342,P=0.016), both have statistical significance.Conclusions1. In mitral valve calcification rheumatism organization membrane, bFGF and BMP2expression rate was higher than normal tissue and the rheumatism mitral valve calcification membrane organization, bFGF and BMP2expression is a significant positive correlation, explain both in rheumatism mitral valve film the happening of the disease development have synergy.2. In mitral valve calcification rheumatism organization membrane, bFGF and BMP2express rises and the lesion calcification degree are concerned, and has nothing to do with age, sex, Pathology classification.
