ObjectiveTo investigate the roles of protein kinase C(PKC)a and K-Ras protein in.the occurrence and development of colorectal cancer.MethodsImmunohistochemistry method was used to detect the PKC-a and K-Rasprotein expression in76colorectal adenocarcinoma,15colorectal adenomatissue and10normal colorectal mucosa, Western Blot method was usd todetect the PKC-a and K-Ras protein expression level in30colorectal cancertissues and10adjacent mucosa,their relationship and correlation with clinicalpathological features of the tumor were analyzed.Results1、Immunohistochemical results: In normal colorectal mucosa、adenomaand colorectal adenocarcinoma,in the positive expression rate of PKC-a were80%(8/10),53.3%(8/15),35.5%(27/76); the positive rate of K-Ras were,10%(1/10),66.7%(10/15),68.4%(52/76) PKC-a in colorectal cancerpositive expression rate was lower than that in normal mucosa (P <0.05),while the positive expression of K-Ras in colorectal carcinoma and adenomawere significantly higher than that in normal mucosa (P <0.05). The PKC-aand K-Ras expression in colorectal carcinoma and the clinical pathological grading, with pathological grading and increase. PKC-a expression ratedecreased gradually, and the K-Ras increase; both the expression and gender,age, lymph node metastasis and Dukes stage (P>0.05) are independent. ThePKC-a and K-Ras protein expression was negatively correlated (r=-0.930,1<R <0).2、Western Blot results:PKC-a protein in colorectal cancer expression levels(0.1044±0.0274) is lower than the corresponding adjacent gastric mucosa(0.5775±0.0377)(P <0.05),PKC-a expression level in the different degree ofdifferentiation between there was a significant difference (P <0.05), K-Rasprotein in colorectal cancer expression levels (0.6004±0.0386) higher than thecorresponding adjacent gastric mucosa (0.0629±0.0066)(P <0.05), K-Rasexpression level in the different degree of differentiation between there was asignificant difference (P <0.05), the results of immunohistochemical findingsconsistent with.Conclusion1, K-Ras protein in colorectal cancer high expression organization, and theorganization of the cancer and differentiation degree relevant, explain theoriginal cancer gene K-Ras may and colorectal cancer development process,and are relevant to a certain extent hints of the tumor after the more.2, PKC-a in colorectal cancer tissue of the lower expression, and with thecancerous tissue differentiation degree will decrease, PKC-a gene that caninhibit tumor occurs, and the differentiation of the tumor levels.3, PKC-a and K-Ras protein in colorectal cancer negatively correlated to theexpression, tip the percentage may be colorectal cancer development disorderprocess one of the mechanism.
