ObjectiveTo observe the effect of the acellular peripheral urethralrepaire the defect of rats and study the immunere sponse ofchemical extraction on th immunogenicity of urethralisograft. In order to develop ideal substitute materials.MethodsThe number of urethrals are20which from SD rats. The length of theurethrals are1.5-cm-long.The urethrals were treated with Triton-100solutionand sodium deoxycholate which reserve the ECM in order to construct thetissue engineer urethrals.10Wistar rats are made into the model of urethralsdefect,and then cured by the ECM of urethrals which from the SD rats.Afterthree weeks the10Wistar rats are made the detections ofdynamia-motivation;electrophysiology and pathology.And then10Wistar ratsare also made into urethral defect and then cure by variant urethral,the rats aretaken into control group.At the same time10Wistar rats are cured by autourethral which are taken into control group.10SD rat urethrals after treatedwith Triton-100solution and sodium deoxycholate bury into Wistar hypo.10SD rat urethrals also bury into Wistar hypo are taken as control group.ResultsAfter3weeks the handle group compare with the auto control group.thedifference between VP;BB;RUV have no significance.(P>0.05).Comparewith variant control group the difference have significance(P<0.05).The result of VG stain shows that the muscle fiber of handle group to line up in order, theregenerate muscle fiber to enter in far-end urethrals.The result of auto controlgroup shows that the muscle fiber to line up in order and the regeneratemuscle fiber to enter in far-end urethrals. The result of variant control groupshows that there have no muscle fiber regenerate.6weeks Acellullar urethralbury handle group compare with control group the difference havesignificance.The wound healing well,H-E stain shows that Acellullar urethral toassimilate by degrees,in stead of smooth muscle and inequality of size bloodvessel enter in far-end urethrals.The result of the control shows that Acellullarurethral not to assimilate by degrees. The result of electrophysiology showsthat stimulate S1nerve can receive the motion evoked potential at far-endurethrals.Auto control group:delitescence1.63±0.48ms,amplitude6.12±0.68mv.Handle group delitescence2.34±0.17ms amplitude2.54±0.22mv. variantcontrol group shows that there is not electromyologram.ConclusionThe acelluldar urethral remove the immunogenicity ingredient.It has themerits of immunogenicity feeble and cell relic remove fully and urethral remainfully.The tissue engineer urethral can repaire1.5-cm-long urethral defect,andhave lower complication rate and has favourable mucouscoat. The acelluldarurethral can form normal urethral construction.
