| ObjectiveTo discuss the impacts of different doses of alcohol on vascular endothelial function, blood lipid and blood glucose of rat, further improve the awareness of drinking impacting the cardiovascular disease, Provide the experimental basis to guide people to a scientific and rational drinking, and to improve thequality of people’s lives.MethodsOrdinary60male SD rats,which were6-8weeks and180-220g, were randomlyallocated into four groups: high dose group (group A), medium dose group (group B),low dose group (group C) and the blank contrast group (group D), each group had15rats. Ethanol feeding to establish an animal model, after the rats were fed alcoholfor8weeks, determination of serum nitric oxide (NO), angiotensin II (Ang II), totalcholesterol (TC), triglyceride (TG), high density lipoprotein (HDL-C), low densitylipoprotein (LDL-C)and blood sugar, and nitric oxide synthase (total NOS,endothelial of eNOS, inducible the type iN0S) activity, and flow cytometer was usedto measure the level of circulating CD31+/CD42-EMPs in each rat, and comparedthe relationship between them and the dose.Results(1) Serum NO, NOS, eNOS, iNOS and AngⅡlevels: Comparing with group D,group A and B: NO (19.89±1.34VS10.75±0.90,15.84±0.89), NOS (31.87±1.46 VS25.05±0.90,27.99±0.99), and eNOS (11.02±1.88VS2.13±1.76,6.84±1.49)decreased, Ang II (821.31±91.37the VS1329.58±65.10,1256.8±100.90)increased, the difference was significant (P <0.05); group C: NO (19.89±1.34theVS20.04±1.44), NOS (31.87±1.46the VS32.63±1.37), eNOS (11.02±1.88theVS12.28±2.05) increased, and Ang Ⅱ (821.31±91.37the VS880.46±73.41)decreased, the difference was no significant (P>0.05); Comparing with groupB,C,D,group A: iNOS in increased (22.92±1.22VS21.15±1.32,20.34±1.01,20.85±0.75), the difference was significant (P <0.05).(2) EMPs levels: Compared with group D, the level of circulating EMPs waselevated in rats of group A and B(1426±108VS2497±155,1976±122), thedifference was significant (P <0.05); there was no obvious change in group C (1426±108VS1407±104), the difference was not significant (P>0.05).(3) Serum blood sugar, TC, HDL-C, LDL-C, TG levels: Comparing with group D,group A and B: Glucose (5.33±0.96the VS6.89±0.16,5.21±0.16) and TC(1.38±0.06VS2.21±0.13,2.01±0.17) increased, the difference was significant (P<0.05), Group C glucose (5.33±0.96VS4.67±0.18) decreased, and increase inTC (1.38±0.06VS1.40±0.56), no significant difference (P>0.05); Comparing withgroup D, group A, B and C HDL-C (0.88±0.07VS1.35±0.09,1.40±1.00,1.41±0.09) increased, the difference was significant (P <0.05); TG and LDL-C betweenthe four groups to change the difference was not significant (P>0.05).Conclusions(1) Small doses by gastro feeding alcohol has a protective effect on rat vascularendothelial cell function, and which can increase serum levels of HDL-C content;(2) High and medium dose by gastro feeding alcohol are harmful to the functionof vascular endothelial cells in rats, which can increase blood sugar levels, affectblood lipids,and shows a obvious dose-effect relationship;(3) Different doses of alcohol plays different role of vascular endothelial cells,which related to the EMPs level and changes in activity of NOS (eNOS and iNOS). |
