| Objective:To observe the immunotherapeutic effects of dendritic cells vaccine pulsedwith the rat pancreatic carcinoma cell lysate on rat with pancreatic carcinoma.Methods:Rat pancreatic cancer cells injected into SD rat pancreatic capsule, theformation of tumor, and then chopped tumor and implanted in rats with the same axillariessubcutaneous, set up nude model; rat pancreatic cells were repeatedly frozen and thawed4times,then extracted from lysates which were cultured for72hours with the SD rat bonemarrow-derived DC to produce DC vaccines. According to DC pulsed, groups were dividedinto DC vaccines groupã€DC groupã€Control group and lysate group,per group five, thenwere injected into nude body. We measured the tumor length and diameter every two days, atotal of8times and observed the survival time of rats, to study their impact on pancreaticcancer nude rat immune treatment.Results: With the time,the tumor size of DC vaccine group decreased; with the time,the tumor size of DC groupã€lysate groupã€control group were increasing trend; DC vaccinegroup and control group differences between the tumor size has statistically significance (P<0.05); DC groupã€lysates group and control groups between the tumor size was nosignificant difference (p>0.05); DC vaccine group and DC group〠lysates group differencesbetween the tumor size has statistical significance (P <0.05).DC vaccine group survival periodof rats is(55.6±9.4)d,significantly longer than other groups(P<0.01).Conclusion: Rat pancreatic cancer cells lysate-pulsed dendrtic cells vaccines can inducea specific and effective immune response against pancreatic carcinoma cell implanted in rat. |
