| In this paper, the quality control method of one of the main raw material of the Tanreqing injection, honeysuckle was researched, including three methods, respectively, is a test of multi-assessment method for the determination of the six kinds of organic acids in the honeysuckle medicinal compounds content, quality control standard extract of honeysuckle herbs, honeysuckle medicine quality control methods based on near infrared similarity matching. The results indicated that all the three methods honeysuckle herbs can be used for quality control.A method of quantitative analysis of multi-components by single marker (QAMS) for quality control of Lonicerae Japonicae Flos was established. The contents of chlorogenic acid (CA) and caffeic acid (CfA) were determined, and the relative correction factors (RCF) of other organic acids were calculated, which were used for the indirect determination. The RCFs for the neochlorogenic acid (NCA),3,4-O-dicaffeoylquinic acid (3,4-DCA),3,5-O-dicaffeoylquinic acid (3,5-DCA), and4,5-O-dicaffeoylquinic acid (4,5-DCA) were5.498,5.733,2.345,2.390(to CA) and3.923,4.099,1.698,1.714(to CfA), respectively. The established method was validated in different laboratories with different high performance liquid chromatography (HPLC) instruments and different chromatographic columns; the result indicated that the reproducibility was satisfactory. There was no significant difference between the established QAMS method and external reference method (P>0.05). The established QAMS method can be used for simultaneous determination of6organic acids as quality control of Lonicerae Japonicae Flos with easy available standard substances.The high price of the reference substances is an obstacle for the HPLC analysis of Lonicerae Japonicae Flos. To solve the questions, a new method based on the Standard Reference Extract (SRE) was proposed. In the study, the extract of Lonicerae Japonicae Flos was calibrated, and the long term stability was investigated. The SRE was used for the preparing of different concentrations solutions. The calibration profiles were established using the solutions and6organic acids were determined. T-test was used for the comparison of the determination results via reference substances and SRE, and the results demonstrated that there is no significant difference between the two methods. The presented method can be used for the quality control of Lonicerae Japonicae Flos, and has special reference to resolve similar problems.In our research,20samples were used as standard samples, and the similarity matching model was established with Gram-Schmidt algorithm after the spectra pretreated with "MSC+1st D+S-G (7,3)" combination method. The Similarity Matching Values (SMVs) of the other10High-quality samples and126samples f bought in the market were calculated and the everage SMVs were98.8%and85.76%, respectively. Therefore, to ensure the consistence of the Lonicerae Japonicae Flos from different batches,90%is taken as the SMV threshold value for the quality test before the raw material purchase.Experiments show that all the three detection methods above mentioned can be used for the quality control Lonicerae Japonicae Flos. |
