Objective:Our purpose was to investigate the effect of promote nerve regeneration factorscompound(N6) in spinal cord injury repair through a proteomics approach and obtain thepotential protein that was induced by N6and may be involved in the spinal cord repairprocesses.Methods:The experimental rats were randomizatized into three groups,namely sham, SCI,SCIN6-treated groups.Spinal cord injury was induced by NYU compactor (New YorkUniversity).Immediately after injury,the SCI N6-treated rats were treated with N6throughsubarachnoid space.Animals were sacrificed at72h following surgery and the spinal cord ofthe contusion epicenter were dissected and stored in liquid nitrogen for protein extraction later.The BBB open field score was used to evaluate the hind limb function of the remainingrats.The study of proteomics: We extracted the protein of the spinal cord and obtained theimages of2-DE. The images were analysised and the differential spots were cutted fromSDS-PAGE and were measured by MS.Result:We evaluate the BBB score of group sham,group SCI,group SCI-N6treated. We couldfound that the difference between SCI and SCI-N6treated were significant at2、3、4weeks.This result showed that N6has effect in thereopying spinal cord injury.From theexperiment,we found about42different spots,and cutted off34spots from SDS-PAGE andqulified27spots with MS.The spots which were not measured were qualified throughMALDI-TOF-TOF,and found2proteins.In the experiment,we qualified29spots in the end and these spots included24proteins.conclusion:N6can promote the hind limb locomotion of the rats induced by spinal cord injury.Fromproteomics we can found differential proteins between group sham and group SCI,theseproteins may play an important role in spinal cord injury pathogenesis.The differential proteinsbetween group SCI and group N6may explain the function of N6in treating spinal cord injuryand provide evidence for further treatment.
