| Objective Observe microvascular Notchl and the expression of VEGF and XueFuZhuYu Decoction intervention effect in rats after acute myocardial ischemia, explore XueFuZhuYu Decoction mechanism of protecting myocardium and its effect on promoting angiogenesis under the perspective of Notch/Delta signal pathwaysMethods Using SD rats,whose coronary artery of left anterior descending branch have been ligated, to establish acute myocardial ischemia model, then randomly divided them into XueFuZhuYu Decoction group7d,7d control group, model group and the heart of musk pill group7d,14d and14d XueFuZhuYu Decoction group and control group, the model14d and14d of musk heart pill group. Taking medicine by lavaging every30minutes,XueFuZhuYu Decoction group and the heart of musk pill keep pumping corresponding drugs every day, control group and model group shipped amount of distilled water. In7d,14d killing animals.Observing myocardial morphology change by HE staining,ischemic myocardial vascular endothelial cells, and the ischemic myocardial tissue micro vascular Notchl, vascular endothelial cell growth factor (VEGF) protein expression by immunohistochemical method.Results1. Myocardial t i ssue morphologi cal changes:control group:the arrangement of myocardial cell nuclear rods, visible horizontal grain, and14d groups of some cells with mild edema, interstitial cell distribution is uniform, rich in capillaries,7d group no inflammatory cell infiltration,14d group with a small amount of inflammatory cell infiltration; Model group:the disordered arrangement of myocardial cells, the cell edema, degeneration, fracture, interstitial broadening, vascular congestion, visible more inflammatory cells infiltration;Heart of musk pill group:myocardial cells arranged disorder, part7d group cell edema,14d group with cell rupture, compared with model group, myocardial cell degeneration degree of ease; Interstitial small amounts of inflammatory cell infiltration; XueFuZhuYu Decoction groups:myocardial cells arranged rules in the model group, mild edema, degeneration was significantly reduced;7d quantity of inflammatory cell infiltration in group stroma and14d groups of cells interstitial distribution is uniform, can be found in a small amount of inflammatory cells infiltration.2. The ischemic myocardial microvascular density (MVD):7d group:MVD increased compared with control group, model group (P<0.05), the treatment group were significantly increased (P<0.01); Compared with model group, the heart of musk pill group increased (P<0.05), the XueFuZhuYu Decoction group increased significantly (P<0.01);14d group:compared with control group, model group and treatment group were significantly increased (p<0.01); Compared with model group, the heart of musk pill group increased (P<0.05), the XueFuZhuYu Decoction group increased significantly (P<0.01); XueFuZhuYu Decoction group14d group than7d had a tendency to rise, but without statistical significance.3. The ischemic myocardial tissue microvascular Notch1protein expression:compared with control group, model group and treatment group were significantly increased (p<0.01); Compared with model group, treatment group were significantly increased(P<0.01); XueFuZhuYu Decoction group14d group than7d had a tendency to rise, but without statistical significance.4. The ischemic myocardial tissue m i crovascular VEGF protein expression:7d groups:model group and control group comparison, MOD value increases (P<0.05); Treatment group compared with control group, were significantly increased (p<0.01); Treatment group compared with model group were significantly increased meaning (P<0.01);14d groups:model group and treatment group were significantly increase, there are significant difference compared with control group statistically significant (p<0.01); Compared with model group, treatment group were significantly increased (P<0.01); XueFuZhuYu Decoction group14d group than7d had a tendency to rise, but without statistical significance.Conclusion XueFuZhuYu Decoction can act against myocardial ischemia injury, this action by reducing the number of myocardial cell apoptosis, adjust Notch/Delta signal pathway, raised the ischemic myocardium area Notchl proteins and VEGF protein expression, and promote the new angiogenesis. |
