Demands for such products have been proved strong with the increase of peoples demands for higher living quality because it has the same effectiveness as Cordyceps sinensis and much cheaper. In recent years, people’s benefits have been damaged heavily because the management of health products market is rather disorderly, business people prioritize profit above all else, national rules and regulations on this type of products are not perfect and there are no legal quality standard to follow. So it is extremely urgent to strengthen the examination of the composition, efficacy and security for such products and set standards.Objective The purpose of this subject is achieve quality control for A Hepialidae hirsutella sinensis powder capsule by purifying SOD and determining its enzymatic activity, separating and determining rapidly nucleotide efficacy component and performing routine examination in such health food products.Methods (1) Ultraviolet spectroscopy has been used to optimize the process of extraction, separation and purification and choose suitable method to determine SOD activity in Hepialidae hirsutella sinensis powder capsule by comparing to T-SOD test kit method. The method will be a guideline to control the quality of A capsule.(2) HPLC-PAD has been used to optimize the extraction and separation conditions of nucleotide in A capsule and determined its limit of detection, then established the HPLC method for A capsula. The qualitative data of nucleotides in A capsule obtained by LC-MS will be a guideline to control the quality of A capsule.(3) Gas chromatograpHy, atomic absorption spectrometry and atomic fluorescence spectrometry have been used to do routine examinations about Hepialidae hirsutella sinensis powder capsule. Results (1) The method of extracting, separating and purifying the SOD in Hepialidae hirsutella sinensis powder capsule and determining enzyme activity has been established. The method applys modified Marklund method to determine the activity of crude enzyme. It extracts SOD from capsule with ultrasonic shaking, uses the pHospHate buffer of pH=7.0as extraction solvent and the extraction time is60min. This method has good reproducibility.(2) HPLC has been established to determining the nucleosides in Hepialidae hirsutella sinensis powder capsule rapidly. The extracting method is ultrasonic shaking, the extraction solvent is purified water, the extracting temperature is40℃and the extracting time is60min.The peaks of four nucleotides in samples could be separated by Shim-pack CLC-ODS column, methanol-water as mobile pHase and gradient elution. The methodology study for this method is great and this method is appropriate for content determination of fresh artificial cultivation cordyceps samples, lyopHilized paecilomyces bepiali chen and nucleotide in conidial powder.(3) HPLC-MS has been established to determining the nucleosides in Hepialidae hirsutella sinensis powder capsule rapidly.(4) General inspections about Hepialidae hirsutella sinensis powder capsule all meet relative requirements for health food.Conclusion (1) The method of separating and purifying the SOD in Hepialidae hirsutella sinensis powder capsule and determining enzyme activity has been established.(2) HPLC has been established to determining the nucleosides in Hepialidae hirsutella sinensis powder capsule rapidly.(3) HPLC-MS has been established to determining the nucleosides in Hepialidae hirsutella sinensis powder capsule rapidly. |
