Objective:To evaluate local implanted with bovine bone morphogenetic protein(ipads morphogenetic protein BMP) on postmenopausal osteoporosisfracture healing of rats. Looking for new ways for clinical prevention andtreatment of osteoporosis fracture.Methods:Randomly selected80adult female SD rats (3months),surgery willovaries make postmenopausal osteoporosis rat model by dual-energyX-ray absorptiometry (dexa) building successful region.perform48ratsonly, set the contralateral limb in rats as a comparison,the experimentalgroup member selected from the big trochanter is about0.5cm at thefemoral neck implanted with bovine bone morphogenetic protein,controlgroup implanted with bovine serum albumin. In28days after surgery,56days based on anesthesia,detection of pathological observation,CT,dual-energy X-ray absorptiometry (dexa) and universal biomechanics toobserve rat femoral neck bone shape and bone cortical thickness,thechange of bone mineral density and bone strength, and taking the rt-pcrmethod in the detection of local bone tissue VEGF,OPG and RANKLgene expression. Results:1、local implantation of BMP to castration rat femoral neck bonemorphology, bone density and biomechanics of bone impact experiments.(1)Observation in the rat femoral neck bone tissue morphologyobserved in28days after building experimental femoral neck localtrabecular bone integrity,continuity is good;Control group local trabecularbone and only a few of the femoral neck into network incomplete.56days experimental femoral neck bone trabecula density,the rise in theNumbers and complete,and connected into a mesh structure,uniformdistribution; Most of the control group did not see the femoral neck bonetrabecular, medullary cavity, delayed fracture healing.(2)Bone density and biomechanics of bone cortex thicknessdetermination:28days there was no significant difference betweenexperimental group and control group at the femoral neck cortexthickness,BMD and maximum load has significant difference (p <0.05);56days experimental bone cortex thickness increased, bone density,maximum load are increased, there was a significant difference comparedwith control group (p <0.05).2、Implanted cattle BMP femoral neck of castrated rats and localVEGF,OPG and RANKL gene expression results show that the influenceof femoral neck bone tissue by rt-pcr method detection test group ofVEGF,OPG and RANKL gene expression was high, there is statistical significance (p <0.01).Conclusions1、Castration rat femoral neck fracture after local implantation ofBMP can increase the thickness of bone cortex, bone density andbiomechanics strength, can improve the reconstruction of bureau oftrabecular bone, prevent bone tissue destruction.2、Into BMP can promote local bone tissue VEGF,OPG and RANKLgene expression,inhibition of osteoblasts apoptosis may be BMP is themain reason for the treatment of osteoporosis.
