Objective: Aquaporins are expressed in many organs and tissues. AQP1is oneof the most important parts in aquaporin family. It not only participates intransmembrane transprotport of water, but also takes part in the proliferation,differentiation and migration of cells. Currently, result has proved that it is expressedin some tumour cells, such as gastric cancer, brain glioma, rectal cancer, prostatecancer and so on. It is also involved in the differentiation of tumour cells. We want totest AQP1’s expression in non-small cell lung cancer and its other functions, andwould like to know whether AQP1plays an important role in apoptosis of non-smallcell lung cancer in this paper.Methods: We have collected the pathological specimens in operations of lungcancer patients in Shenyang Cancer Hospital from May2011to May2012. Thepatients in our study were not treated by radiation therapy and chemotherapy beforeoperations. They are all adult males and their age was from30to60. Thepathological specimens were diagnosed by doctors and we found the patients whodeveloped lung squamous cell carcinoma and adencarcinoma. There were15specimens of well differentiated squamous cell carcinoma,15specimens of poordifferentiated squamous cell carcinoma,15specimens of well differentiatedadenocarcinoma and15specimens of poor differentiated adenocarcinoma. Thespecimens totaled60. Meanwhile, we got some paraneoplastic tissues from patients.We observed and tested the degree of the differentiation of lung squamous carcinomacells and lung adenocarcinoma cells, the level of expression of AQP1as well as theapoptosis rate of lung squamous carcinoma cells with methods of HE staining andAQP1immunohistochemistry. Then we conducted optical density test of AQP1inthe expression of squamous cell lung carcinoma by IPP6software and analyzed therelation between AQP1and apoptosis of cells by SPSS18.0.Results: The average optical density of AQP1in the expression of well differentiated squamous carcinoma cells is758.44±112.25; the average opticaldensity of AQP1in the expression of poor differentiated squamous carcinoma cellsis487.21±74.32. There is a significant difference between them (P﹤0.05). Theaverage optical density of AQP1in the expression of well differentiatedadenocarcinoma cells is1118.08±46.20; The average optical density of AQP1in theexpression of poor differentiated adenocarcinoma cells is819.61±59.79. There is avery significant difference between them (P﹤0.01). The apoptosis rate of welldifferentiated squamous carcinoma cells is12.04±2.36; the apoptosis rate of poordifferentiated squamous carcinoma cells is7.74±1.03. There is a very significantdifference between them (P﹤0.01). The apoptosis rate of well differentiatedadenocarcinoma cells is28.40±0.89; the apoptosis rate of poor differentiatedadenocarcinoma cells is18.24±0.80. There is a very significant difference betweenthem (P﹤0.01). We obtained the result of the relation between AQP1in theexpression of lung squamous carcinoma cells and the apoptosis rate of cells (r=0.693). There is a very significant difference between them (P﹤0.01). There is apositive correlation between AQP1and the apoptosis of lung squamous carcinomacells. We get the result of the relation between average optical density of AQP1inthe expression of adenocarcinoma cells and apoptosis rate (r=0.727). There is a verysignificant difference between them (P﹤0.01). There is a positive correlationbetween AQP1and the apoptosis of adenocarcinoma cells.Conclusion: AQP1has a high expression in well differentiated squamouscarcinoma cells and has a low expression in poor differentiated squamouscarcinoma cells; The apoptosis rate of well differentiated squamous carcinoma cellsis higher than apoptosis rate of poor differentiated’ squamous carcinoma cells; Thereis a positive correlation between AQP1and the apoptosis of lung squamouscarcinoma cells, and the result suggested that AQP1is involved in the apoptosis oflung squamous carcinoma cells. AQP1has a high expression in well differentiatedadenocarcinoma cells and has a low expression in poor differentiatedadenocarcinoma cells; the apoptosis rate of well differentiated adenocarcinoma cells is higher than that of poor differentiated adenocarcinoma cells; There is a positivecorrelation between AQP1and the apoptosis of lung adencarcinoma cells, and theresult suggested that AQP1is involved in the apoptosis of lung adencarcinoma cells.
