ObjectTo observe the effect of muscle basal lamina(MBL) with neural stem cells(NSCs) and olfactory ensheathing cells(OECs) in spinal cord injury(SCI) repair.MethodThe72SD rats were randomly divided into six groups, the blank control group (group A), the NSCs transplantation group (group B), the OECs transplantion group (group C), the MBL with NSCs transplantation group (group D), the MBL with OECs transplantation group (group E), the MBL with NSCs and OECs transplantation group (group F). All of the groups were made into the spinal cord hemisection model. In group A, the ends of spinal cord hemisection model only were flushed with physiological saline. In group B, the NSCs in the ends were injected. In group C, the OECs in the ends were injected. In group D, the MBL with NSCs was implanted into the ends. In group E, the MBL with OECs was implanted into the ends. In group F, the MBL with NSCs and OECs was implanted into the ends. The survival and migration of the transplantation cells were detected by the immunohistochemistry and immunofluorescence after4and8weeks. In addition, the hind limb function repair was evaluated by Bundle branch block (BBB) score in various time points before and after surgery.Results1. Pathological features of the4weeks after surgery:In group A, there has fibrous connective tissue hyperplasia, spinal cord tissue necrosis and cavity formation. In group B, there has a large cavity in the ends, the NSCs were scarce, the inflammatory response was heavy, the fibrous scar was obvious. In group C, the positive staining of nerve fibers was less and disarranged, the infiltration of inflammatory cells was obvious. In group D, the transplanted cells in the MBL migrated to the head and tail, the host cell has entered into the MBL, the transplanted cells and the surrounding normal spinal cord tissue were fusion. In group E, the OECs formed myelin around the axons, the infiltration of inflammatory cells was not obvious. In group F, the NSCs and OECs grew in MBL, part of the neural stem cells has into neurons.2. The results of the electron microscope:The MBL of group F did not occur autolysis, there has the Nissl bodies of neurons, axons and synapses, but the outer layer of axons appeared nerve demyelination.3. The BBB score:The BBB scores of D, E groups higher than the other groups, and the BBB scores of D group higher than E group. This has significant differences (P<0.05). The BBB scores of G group higher than A,B,C,E groups, but there was no significant difference in the F group and D group (P>0.05)Conclusion1. Muscle basal lamina can promote growth of neural stem cells along its lumen, and promote host cells crawling in its lumen. It can reduce the local inflammatory response. The method of muscle basal lamina with NSCs or/and OECs for repair spinal cord is better than the simple cell transplantation.2. In group F, there are more nerve cells in its muscle basal lamina than group E. Therefore, this is conducive to form more axons, synaptic connections and signal transduction pathways. However, due to these new axons presented nerve demyelination, may greatly limit the conduction of nerve signals.3. In group F, OECs have some help to induction of neural stem cells, axonal growth and the formation of synaptic connections, but its role is limited.
