| Objective: Observation the dynamic change of caspase-8expression and researchthe control mechanism of cell apoptosis in rat liver regeneration process.Methods:75healthy male SD rats were divided into3groups. Partial hepatectomygroup, n=35.We do70%of liver resection surgery to the rat after10%chloral hydrateanesthesia. Sham-operated hepatectomy group, n=35. Turn the liver only, no liverresection. Normal control group only5rats. Do not do any processing. Partialhepatectomy group and Sham-operated hepatectomy group each is divided into sevengroups. We put the rat to death after operated at3h,6h,24h,3d,7d,11d and14d. Cutout the residual liver, and weight. Normal group directly for the liver. Histologicalprocessing, Paraffin embedding, Sectioning. Do H-E staining and immunohisto-chemical staining. Read the liver cell morphological changes at different time point.Detection the expression and distribution of caspase–8at different time points.Results:①Liver regeneration degree. The liver regeneration degree at3h after thesurgery of liver regeneration is0.15±0.02. The liver regeneration degree at6h is0.17±0.01. The liver regeneration degree at24h is0.21±0.02. The liver regenerationdegree at3d is0.52±0.04. The liver regeneration degree at7d is0.82±0.04. The liverregeneration degree at11d is0.83±0.03. The liver regeneration degree at14d is0.85±0.02. The trend of the change is started to rise at24h, reach the top at7d.②Index of liver regeneration. The index of liver regeneration at3h after the surgeryof liver regeneration is0.99±0.10. The index of liver regeneration at6h is1.09±0.16.The index of liver regeneration at24h is1.12±0.03. The index of liver regeneration at3d is2.15±0.08. The index of liver regeneration at7d is3.30±0.07. The index of liver regeneration at11d is3.33±0.17. The index of liver regeneration at14d is3.35±0.09.The trend of the change is started to rise at24h, increased dramatically at3d, reachthe top at7d.③H-E staining. The normal group. The structure of the hepatic lobule iscomplete. There are radial arrangement liver cell line around central venous. Thevolume of the liver cells is normal. There are no obvious inflammatory cellsinfiltration. The sham-operated hepatectomy group. The structure of the hepaticlobule is complete. There are radial arrangement liver cell line around central venous.The volume of the liver cells is normal. There are no obvious inflammatory cellsinfiltration. Did not see clearly mitosis. Normal group and control group rarely seeliver cells nuclear division phase. Partial hepatectomy group. The liver cells edemaand transgender is lighter at3h after the surgery of liver regeneration. Inflammatoryreaction is not obvious. The liver blood sinus is broaden little. The swelling of the cellis clearly at6h compared with the previous. The liver blood sinus is furtherbroadening. There are more red blood cells in liver blood sinus. The structure of thehepatic lobule is complete. The swelling of the cell is reduced at24h compared withthe previous. But the blood sinus clearance is further increase compared with theprevious. There are more red blood cells in blood sinus. And dual-core liver cellsincreased in. At3d after the operation, liver cells increased obviously and the size ofthe nucleus is different. There are cavitation at the cytoplasm. There are dual-coreliver cells and mitosis around portal area. The blood sinus interval fuzzy after7d. Thedual-core liver cells increased. The phenomenon of cavitation is more earlier. After11d most liver plate arrangement is neat, liver cells and nuclear size is differ, dual-coreand mitosis decreased.14d after hepatic the lobule structure is complete, liver sinus isobvious, most liver plate arrangement is neat, the degree of liver cell and the nuclearsize is differ, dual-core and mitosis decreased. The phenomena of rare mitotic atnormal control group and control group are little. The phenomena of rare mitotic ismost at3d. At11d,14d rarely see mitosis.④The immunohistochemical staining. The caspase-8protein mainly distributed in thenucleus and its positive product is tan. The percentage of positive cell at normal control group is3.51±2.38%. The percentage of positive cell at sham-operatedhepatectomy group at3h is10.34±5.32%. The percentage of positive cell atsham-operated hepatectomy group at6h is12.53±4.56%. The percentage of positivecell at sham-operated hepatectomy group at24h is14.23±4.13%. The percentage ofpositive cell at sham-operated hepatectomy group at3d is6.12±4.67%. Thepercentage of positive cell at sham-operated hepatectomy group at7d is4.65±3.75%,The percentage of positive cell at sham-operated hepatectomy group at11d is3.53±4.26%. The percentage of positive cell at sham-operated hepatectomy group at14d is3.46±5.37%. Compared3h,6h,24h,3d,7d groups with normal group, the P isless than0.05. The difference have statistically significant. Compared11d and14dgroups with normal group, the P is more than0.05. The difference have no statisticallysignificant. The percentage of positive cell at partial hepatectomy group at3h is26.74±4.59%. The percentage of positive cell at partial hepatectomy group at6h is30.58±3.58%. The percentage of positive cell at partial hepatectomy group at24h is32.16±4.36%. The percentage of positive cell at partial hepatectomy group at3d is53.19±5.43%. The percentage of positive cell at partial hepatectomy group at7d is70.36±7.69%. The percentage of positive cell at partial hepatectomy group at11d is50.37±4.38%. The percentage of positive cell at partial hepatectomy group at14d is45.25±7.67%. Compared seven partial hepatectomy groups with normal group, the Pis less than0.05. The difference have statistically significant. Compared seven partialhepatectomy groups with seven sham-operated hepatectomy groups, the P is less than0.05. The difference have statistically significant. The trends of caspase-8expressionat partial hepatectomy group begin to rise at3h, to7d reach the mountain and beginto decline, while to14d were significantly higher than the sham-operatedhepatectomy group and normal control group. The trends of caspase-8expression atsham-operated hepatectomy group begin to rise at3h, to24h reach the mountain andbegin to decline, at11d recovered to normal level.Conclusion: Caspase–8is the perform enzyme in early apoptosis. There is amolecular regulation mechanism in cell apoptosis after most of liver resection. Theactivities of cell apoptosis in early may be caused by the surgical stress response. The cell apoptosis continue to increase in the late may be related to the death of liverregeneration and the regulation of liver tissue remodeling. |
