| Objective:To explore the effect of Wen Yang Zhen Shuai granules on chronic heart failure model of rabbit cardiac function, JNK, phosphorylation of JNK (p-JNK) expression, JNKmRNA influence the transcriptional level expression, and provide a theoretical basis for the prevention of chronic heart failure with traditional Chinese medicine.Methods:15rabbits were selected randomly from60rabbits which lived in appropriate environmental conditions and raised adaptability after a week and allocated into normal group, and the remaining45rabbits were allocated into model group. The model group was injected with Adriamycin(ADR)(1mg/kg, mixture of1.0mg/ml ADR in0.9%NaCL injection solution) through ear marginal vein for4weeks to construct the experimental rabbit model of chronic heart failure(CHF). After4weeks, all the experimental rabbits model were checked by echocardiography. Based on the principle of random,13rabbits were selected from43experimental rabbits model which survived after the injection of Adriamycin and allocated into the4weeks group of experimental rabbit model. Then this group would be put to death and separated out their heart which will be measured the total proteins of JNK expression and the protein expression of JNK phosphorylation level (p-JNK) and quantificationally analysised the expression of JNKmRNA’s transcription. The remaining30experimental rabbit which had been injected Adriamycin would be divided into5groups by the principle of random according to the result of echocardiography and cardiac function, and named for model control group(A)ã€the positive control group (B)ã€the low dose group of Wen Yang Zhen Shuai granules(C)ã€the middle dose group of Wen Yang Zhen Shuai granules (D)ã€the high dose group of Wen Yang Zhen Shuai granules (E). Excepted the normal control group were injected with saline, the remaining5groups were injected with Adriamycin for another4weeks. Meanwhile, normal control group and model control group were lavaged with physiological saline; the low dose group of Wen Yang Zhen Shuai granulesã€the middle dose group of Wen Yang Zhen Shuai granulesã€the high dose group of Wen Yang Zhen Shuai granules were lavaged with Wen Yang Zhen Shuai granules, positive control group were lavaged with enalapril. After the experiment, the survived rabbits were checked by echocardiography, and put it to death quickly and separated out their heart to test the total proteins of JNK expression and the protein expression of JNK phosphorylation level (p-JNK) and quantificationally analysised the expression of JNKmRNA’s transcription. Results:1.Comparison of cardiac function:constructing the model4week s later, experimental rabbit left heart function decrease obviously than normal control group and the difference has statistical significance (P<0.05).8weeks later, compared with normal control group, the left ventricular function of control group decreased significantly. Positive control group and Wen Yang Zhen Shuai low dose group, Wen Yang Zhen Shuai middle dose group, Wen Yang Zhen Shuai high dose group were improved to varying degrees and the difference has statistical significance (P<0.05), compared with control group.Compared with Positive control group and Wen Yang Zhen Shuai low dose group, the left ventricular function of Wen Yang middle and Wen Yang high dose group was improved obviously, the difference has statistical significance (P<0.05). The difference between Wen Yang Zhen Shuai low dose group and positive control group has no statistical significance (P>0.05). And the same to the difference between Wen Yang Zhen Shuai middle dose group and Wen Yang Zhen Shuai high dose group(P>0.05).2.JNK protein expression:There was no obvious change in each group and the difference has no statistical significance (P>0.05).3.The expression of JNK protein phosphorylation:4week model group model control group, positive control group and Wen Yang Zhen Shuai low dose group, Wen Yang Zhen Shuai middle dose group, Wen Yang Zhen Shuai high dose group are all higher than normal control group,the difference has statistical significance (p<0.05). And the p-JNK of drug intervention group decrease than the control group of the same period, the difference has statistical significance (p<0.05). The difference between Wen Yang Zhen Shuai high dose group and positive control group has no statistical significance(p>0.05) and the same to the difference between Wen Yang Zhen Shuai dose group and Wen Yang Zhen Shuai middle dose group(P>0.05).4. JNKmRNA expression:build model before and after JNK relative expression of mRNA there was no statistically significant difference between the groups (P>0.05).Conclusion:Results of this trail suggest that Wen Yang Zhen Shuai granules and enalapril could cut the pathway of signal transduction of JNK expression level, and improve left ventricular function, and inhibit cell apoptosis, and improve ventricular remodeling, to prevent heart failure effect. |
