Objective: Through investigating the expression as well as theclinicopathological characteristics between the proteins of SMYD2and PTTGin colorectal carcinoma, it was explored the correlations between the positiveexpression rate of SMYD2and PTTG proteins and the invasion andmetastasis in colorectal cancer, and offered a clinical basis for the diagnosisand prognosis.Methods:43colorectal cancer cases(not with radiotherapy andchemotherapy) that were underwent surgery(including23Rectum cancer,6Descending colon cancer,5Transverse colon cancer,9Ascending coloncancer;13well-differentiated adenocarcinoma,20differentiatedadenocarcinoma,10poorly-differentiated adenocarcinoma)were collected inAffiliated Hospital of North Sichuan Medical College from July2012toJanuary2013. Immunohistochemical staining methods was used to detect theexpression of the SMYD2and PTTG protein in all the colorectal carcinomatissue.Results:1SMYD2protein was discriminated the nucleus from the cytoplasm incolorectal cancer cells. The positive expression rate of SMYD2protein in thecancer tissue, the intestinal lymph node tissue, and the mesenteric artery rootlymph node tissue was86.04%,65.11%and30.23%, and that in the canceradjacent tissue and the normal tissue was20.93%and16.27%.2PTTG protein was overexpressed in the cytoplasm in colorectal cancercells, and the positive expression rate of the protein in the cancer tissue, the intestinal lymph node tissue, and the mesenteric artery root lymph node tissuewas88.37%,33.25%and25.58%, and that in the cancer adjacent tissue andthe normal tissue was13.95%and11.62%.3The positive expression of SMYD2protein in colorectal cancer was nocorrelation with gender, age, the tumor location, the lymph node metastasisand the TNM staging(P>0.05); it was positively correlated with the serumCA19-9, the result of the fecal occult blood(immune) and the depth ofinvasion (P<0.05), and was negatively correlated with the differentiation ofcancer cell(P<0.05).4In colorectal carcinoma the positive expression of PTTG protein wasno correlation with gender, age, the tumor location, the depth of invasion, thelymph node metastasis, the TNM staging and the serum CA19-9(P>0.05); itwas positively correlated with the result of the fecal occult blood(immune)(P<0.05), and was negatively correlated with the differentiation of cancercell(P<0.05).5It was confirmed significant elevation of SMYD2in all the differenttissues that of the cancer tissue, the cancer adjacent tissue, the intestinallymph node tissue, the mesenteric artery root lymph node tissue and thenormal tissue (P<0.001). Pairwise comparion of the expression in varioustissues: Comparative analysis of the cancer tissue was distinguishedsignificantly from the other tissues such as the intestinal lymph nodetissue(P<0.001); And there was statistically significant that the intestinallymph node tissue was differented from the cancer adjacent tissue, themesenteric artery root lymph node tissue and the normal tissue; But there wasno statistically significant that the normal tissue was dissimilared from thecancer adjacent tissue and the mesenteric artery root lymph node tissue, as well as the intestinal lymph node tissue from the mesenteric artery root lymphnode tissue(P>0.05).6In all the different tissues there was a significant elevation of PTTG incolorectal carcinoma that of the cancer tissue, the cancer adjacent tissue, theintestinal lymph node tissue, the mesenteric artery root lymph node tissue andthe normal tissue(P<0.001). Pairwise comparion of the expression in varioustissues: Comparative analysis of cancer tissue was distinguished from theother four tissues and was statistically significant (P<0.001). And there wasstatistically significant that the intestinal lymph node tissue that wasdifferented from the cancer adjacent tissue from cancer tissue and the normaltissue(P<0.05). There was no statistically significant between the theintestinal lymph node tissue and the mesenteric artery root lymph node tissue(P>0.05), there was no statistically significant between the adjacent tissuefrom the cancer tissue and the normal tissue (P>0.05).7In colorectal carcinoma it was showed by using Spearman correlationanalysis that there was positive correlation between the proteins of SMYD2and PTTG in cancer tissue(r=0.449,P=0.001), but no correlation in the rest ofthe other tissues (P>0.05).Conclusion:1SMYD2and PTTG proteins’ expression in colorectal carcinoma wasmainly located in the nucleus and cytoplasm of cancer cells.2In colorectal carcinoma there were differential positive expression rateof SMYD2and PTTG proteins in the cancer tissue, the cancer adjacent tissue,the intestinal lymph node tissue, the mesenteric artery root lymph node tissueand the normal tissue.3SMYD2and PTTG proteins’ expression in colorectal carcinoma was positively correlated.
