| Part I The influence of Azithromycin and Dexamethasone on the ICAM-1and TNF-asecretion by A549cells stimulated with Aspergillus fumigatus.Objective:To observe the level of ICAM-1and TNF-a by A549cells stimulated with Aspergillus fumigatus secretion and the influence of Azithromycin and Dexamethasone on the ICAM-1and TNF-asecretion.Method:0.5/2.5mg Azithromycin and0.5/2.5mg Dexamethasone were used to coculture with A549cells differently. After4h and24h the supernatant was harvested and the levels of ICAM-1and TNF-a secreted by A549cells were measured using ELISA.Results:ICAM-1and TNF-asecreted by A549cells were significantly increased; With the extension of time, the release of ICAM-1and TNF-awere increased significantly; Azithromycin and Dexamethasone can significantly inhibit the secretion of ICAM-1and TNF-a, and it is related with the dose of the drug.Conclusions:Aspergillus fumigatus could induce A549cells to release ICAM-1and TNF-a. Azithromycin and Dexamethasone can significantly inhibit the secretion of ICAM-1and TNF-a in A549cells incubated with Aspergillus fumigatus and reduce inflammatory response.Part II Anti-inflammatory of azithromycin and dexamethasone in Aspergillus fumigatus infection and effects on invasion of spore.Objective:To observe the influence of Azithromycin and Dexamethasone on the IL-6and IL-8secretion by A549cells stimulated with Aspergillus fumigatus.Method:A549cells and bronchial epithelial cells were stimulated by Aspergillus fumigatus spore suspension respectively and used to coculture with0.5/2.5mg Azithromycin and0.5/2.5mg Dexamethasone differently. After4h and24h the supernatant was harvested and the levels of IL-6and IL-8secreted by A549cells were measured using ELISA.Results:1.IL-6and IL-8secreted by A549cells stimulated with Aspergillus fumigatus were significantly increased; Azithromycin and can significantly inhibit the secretion of IL-6and IL-8;2.①he secretion of IL-6was decreased in2.5mg/L dexamethasone group than in the0.5mg/L dexamethasone group, P=0.092;②The secretion of IL-6was decreased in2.5mg/L dexamethasone group than in the spores group, P=0.060.③The secretion of IL-8was decreased in2.5mg/L dexamethasone group than in the0.5mg/L dexamethasone group, The diference between the two groups was statistically significan (P=0.010)④The secretion of IL-8was decreased in2.5mg/L dexamethasone group than in the spores group, The diference between the two groups was statistically significan (P=0.017).⑤The secretion of IL-6was decreased in2.5mg/L Azithromycin group than in the0.5mg/L Azithromycin group, P=0.079;⑥The secretion of IL-6was decreased in2.5mg/L Azithromycin group than in the spores group, P=0.062;⑦The secretion of IL-8was decreased in2.5mg/L Azithromycin group than in the0.5mg/L Azithromycin group, The diference between the two groups was statistically significan(P=0.025).⑧The secretion of IL-8was decreased in2.5mg/L Azithromycin group than in the spores group, The diference between the two groups was statistically significan (P=0.025).3.Dexamethasone could enhance the invasiveness of spore and azithromycin can’t.Conclusion:Azithromycin and Dexamethasone can inhibit the secretion of IL-6and IL-8in A549cells incubated with Aspergillus fumigatus and reduce inflammatory response in organs.
