| Background and objectiveOvarian cancer is the highest degree malignant gynecological tumors in female, it is the lowest five-year survival rate of malignant cancer diseases. Besides disease staging, it is closely related to survival rates, and with postoperative chemotherapy drug resistance hased on platinum. At present, it was found Annexin A3protein is the main protein of ovarian cancer cisplatin resistance. The purpose of this experiment is that studies on Annexin A3,TAP73and P53expression in epithelial ovarian carcinoma and their correlation.Materials and MethodsAll samples were collected from the surgical excision ovariectomy from July2011to March2012at Obstetrics and Gynecology of the Second Affiliated Hospital of Zhengzhou University. The experimental group without preoperative chemotherapy, postoperative pathology confirmed for patients with epithelial ovarian cancer; The control group is no significant ovarian lesions, surgical treatment due to other benign diseases requires removal of the ovaries, pathologically confirmed for patients with normal ovarian tissue, and record the relevant clinical information.1. Source of specimens:During July2011to March2012, we collected Patients information and tissue samples with surgical resection of ovarian in the second Affiliated Hospital of Zhengzhou University, All cases neither the preoperative chemotherapy nor postoperative, which were pathologically comfirmed. Including16cases of epithelial ovarian cancer (the experimental group),21cases of normal ovarian(the normal group). The experimental group was treated platinum-based standard chemotherapy after surgery, according to the scores of CA-125determine follow-up experimental groups of disease progression, and divided experimental group to Ovarian cancer resistant group (6cases)and Platinum-sensitive group (10patients). And we Collected groups of patients with intraoperative removal of fresh tissues Samples of corresponding archival paraffin blocks in pathology department are also collected.2. Experimental methods:RT-PCR was used to detect the expression of Annexin A3mRNA, TAP73mRNA and P53mRNA in the two group, and immunohistochemical SP method was conducted to detect the expression of Annexin A3protein, TAP73protein and P53protein in each group.3. Statistical analysis:All dates were analysised by SAS V9statistcal software. t test was used for the date consistent with normality and homogeneity of variance, Rank sum test for the date, which was not consistent with normality and homogeneity of variance. The expression of Annexin A3:RT-PCR results showed that AnnexinA3mRNA in the experimental group was significantly higher than that in the normal group (P<0.05), Annexin A3mRNA in the resistance group was significantly higher than that in the sensitive groups (P<0.05). Immunohistochemical method showed that Annexin A3protein was mainly expressed in the cytoplasm, Annexin A3protein in the experimental group was significantly higher than that in the normal group (P<0.05), Annexin A3protein in the resistance group was significantly higher than that in the sensitive groups (P<0.05).2. The expression of TAP73:RT-PCR results showed that TAP73mRNA in the experimental group was significantly higher than that in the normal group (P< 0.05), TAP73mRNA in the resistance group was significantly higher than that in the sensitive groups (P<0.05). Immunohistochemical method showed that TAP73protein was mainly expressed in the cell nucleus, TAP73protein in the experimental group was significantly higher than that in the normal group (P<0.05), TAP73protein in the resistance group was significantly higher than that in the sensitive groups (P<0.05).3. The expression of P53:RT-PCR results showed that P53mRNA in the experimental group was significantly higher than that in the normal group (P<0.05), P53mRNA in the resistance group was significantly higher than that in the sensitive groups (P<0.05). Immunohistochemical method showed that P53protein was mainly expressed in the cell nucleus, P53protein in the experimental group was significantly higher than that in the normal group (P<0.05), P53protein in the resistance group was significantly higher than that in the sensitive groups (P<0.05).4. The correlation analysis between clinical pathology factors and expression of Annexin A3, TAP73and P53in experiment group:Annexin A3protein expression is not associated with pathological type the tissue differentiation, and surgical-pathological staging. TAP73protein and P53protein expression are associated with the degree of differentiation of ovarian cancer, which expressed in low differentiated than high differentiation (P<0.05), and pathological type, disease staging are no correlation.5. The correlation analysis between Annexin A3, TAP73and P53:Spearman rank correlation analysis showed that the expression of Annexin A3mRNA and TAP73mRNA in epithelial ovarian cancer is positive-correlation (P<0.05,r=0.71494); the expression of Annexin A3mRNA and P53mRNA in epithelial ovarian cancer also are positive-correlation (P<0.05,r=0.74153); the expression of TAP73mRNA and P53mRNA in epithelial ovarian cancer also are positive-correlation (P<0.05,r=0.88165); the expression of Annexin A3mRNA and TAP73mRNA, Annexin A3mRNA and P53mRNA and P53mRNA and TAP73mRNA have no correlation in the resistance group (P=0.3726,0.9087and0.1526), and same to the sensitive group (P=0.6509,0.8389and0.5662). the expression of Annexin A3 protein and TAP73protein in epithelial ovarian cancer are of positive correlation (P<0.05,r=0.9429); the expression of Annexin A3protein and P53protein in epithelial ovarian cancer also are of positive correlation (P<0.05,r=0.8994); the expression of TAP73protein and P53protein in epithelial ovarian cancer also are positive-correlation (P<0.05,r=0.9119); the expression of AnnexinA3protein and TAP73protein, AnnexinA3protein and P53protein and P53protein and TAP73protein have no correlation in the resistance group (P=0.0517,0.6001and0.1321), and same to the sensitive group (P=0.6469,0.6796and0.2339).Conclusions1. The overexpression of Annexin A3in resistance group of epithelial ovarian cancer that indicates Annexin A3is closely related to platinum resistance, and the overexpression of Annexin A3often has poor prognosis in epithelial ovarian cancer.2. The expression of TAP73and P53in the experimental group is significantly higher than that in the normal group (P<0.05), they closely associated with the epithelial ovarian cancer; they expression were associated with the degree of differentiation of ovarian cancer, Poorly differentiated has high expression, it prompt that they are related to prognosis.3. The expression levels of Annexin A3, TAP73and P53has a significant positive correlation in epithelial ovarian cancer, it prompt that they have closely connection. |
