Objective:To investigate the expression and clinical significance of scinderin inepithelial ovarian cancer and to investigate the effect of small interferenceRNA(siRNA)-mediated scinderin silencing on the effect of cisplatin-resistantSKOV3/cDDP cells.Methods:The expression of scinderin in epithelial ovarian cancers, benign ovarianepithelial tumors and normal ovarian tissues were examined by real-time PCR andwestern blot. The correlation between the expression of scinderin andclinicopathologic features were further analyzed.The expression of scinderin gene inSKOV3cells and SKOV3/cDDP cells was determined by real-time PCR. The RNAinterference (siRNA) technique was used to interfere with the expression of scinderinin SKOV3/cDDP cells.The sensitivity of the SKOV3/cDDP-siRNA cells to cisplatinwas detected by MTT assay.The apoptosis rate of SKOV3/cDDP-siRNA cells wasexamined by flow cytometry(FCM).Results:The expression of scinderin was significantly higher in epithelial ovarian cancersthan in normal ovarian tissues and benign ovarian epithelial tumors (P<0.05). Theexpression of scinderin has no significant difference in normal ovarian tissues andbenign ovarian epithelial tumors(P>0.05). The expression of scinderin has acorrelation with histological grades and1ymphatic metastasis of ovarian cancers(P<0.05),but not with age and FIGO stages(P>0.05).The expression of scinderingene in the SKOV3/cDDP cells was higher than that in the SKOV3cells(P<0.05). Scinderin-specific siRNA efficiently down-regulated scinderin expression inSKOV3/cDDP cells(P<0.05). Inhibition of scinderin expression could make the IC50and drug-resistant index of cisplatin decrease, and enhance the sensitivity of the cellto cisplatin.The apoptosis rate was higher in the interference group as compared withthe other two control groups(P<0.05).Conclusion:Expression level of scinderin was significantly higher in epithelial ovariancancer. Ovarian Cancer with high grade and1ymphatic metastasis have higherexpression of scinderin. Scinderin silencing reverses resistance to cisplatin to someextent and enhances the apotosis of SKOV3/cDDP cells.Scinderin is associated withpoor prognosis in patients with ovarian cancer, and it may play an important role inovarian cancer cisplatin resistance, so it could be a biological marker for ovariancancer.
