Objective To investigate the beneficial effects of Ginsenoside Rg1(Rg1) in thetreatment of the experimental rats of cardiac hypertrophy induced by transverseaortic contraction(TAC) and the potential molecular mechanisms; To providesufficient scientific evidence for HIF-1α as a new therapeutic target to therapycardiac hypertrophy, and adequate theory for Rg1as a durg to therapy cardiachypertrophy.Methods Adult male SD rats were conducted in the study; twenty-four ratswhich are managed to present left ventricular hypertrophy (LVH) after TAC wererandomly divided into two groups: the TAC group and the Rg1group. Another12rats which was absence of the aortic artery ligated after analogous operation wasincluded in the Sham group. The administration of Rg1was conducted at10mg/kg/day for4weeks. After that, doppler and hemodynamics was performed toanalysis the cardiac function, the left ventricular mass index (LVMI) were examinedas hypertrophic parameters; left ventricle sections were stained with hematoxylinand eosin for ventricular wall thickness and myocardial microstructure. ThemRNA expressions of ANF, BNP and HIF-1α were detected by Real-timequantitative PCR. The protein expression of HIF-1α and p-AKT was evaluatedthrough Western blot test.Results LVEDD, IVSd, LVPWd and Lvvols determined by echocardiography weresignificantly increased in TAC rats (P<0.05), EF and FS determined by echocardiography were significantly reduced in TAC rats (P<0.05); After treatment with Rg1, LVEDD, IVSd, LVPWdand Lvvols determined by echocardiography were significantly reduced in TAC rats (P<0.05),EF and FS determined by echocardiography were significantly improved in TAC rats (P<0.05).Furthermore, Rg1pretreatment significantly decreased Cardiomyocyttes cross-sectional area inTAC rats (P<0.05). Additionaly, Rg1attenuated the increase in hppertrophic marker expressioncaused by TAC. Data revealed that Rg1reduced the increase of the expression of transcripts ofhypertrophic markers, ANF and BNP, and heightened increased of the expression of transcriptsof HIF-1α in TAC rats. Western blot showed the increased HIF-1α and p-AKT/AKT protenexpression in TAC rats. Rg1augmented the increase of HIF-1α and p-AKT/AKT protenexpression in TAC rats.Conclusions Ginsenoside Rg1attenuates the left ventricular hypertrophy andimproves the cardiac function in the experimental rat model after TAC undertaken.This beneficial effects may be partially attributed to directly activating PI3K/AKTand the increased the HIF-1α expression.
