Bidrectional-fermentation is called the medicianal fungal new type (bidirectioanl) solid-state fermentation engineering,That the active ingredients of Chinese herbal medicines as medicinal matrix to replace the traditional fungal culture (wheat bran, bagasse and other agricultural products).The potency matrix in the fermentation process not only provides such as fungal required carbonitride nutrients,the enzyme produced by the fungus also synthesis or decomposition of the herbs contained in the ingredients on the basis of new ingredients.Achieve increased efficacy, produce new sexual taste, function, and reduce the toxicity of raw herbs. Bidirectional fermentation technology without medicinal fungal secondary metabolites separation, two together, synergistic pharmacodynamic.Trametes robiniophila Murr, the successful development of medicinal fungi. Huai Qi fungal substance, its research and development has achieved a good clinical effect.Rhubarb as commonly used in traditional Chinese medicine, a diarrhea attack plot, detoxification, blood circulation and other effects, is widely used in traditional Chinese medicine clinical treatment of various diseases.Rhubarb used without handled has strong cathartic effect,is easy to cause abdominal pain, nausea and other gastrointestinal reactions.Binding anthraquinone derivatives, known rhubarb purgative ingredients.In order to ease the cathartic effect of rhubarb and gastrointestinal adverse reactions, often by heating or plus accessories concocted into various processed products. Heating can make the anthraquinone decomposition or destruction, so as to ease diarrhea and other effects.The methods and results of this study are as follows:(1)The vaccination of Trametes robiniophila Murr in rhubarb herbs, under growth conditions of Trametes robiniophila Murr, dry discount rate, consumption rate and polysaccharide content change, inspect the adaptability of the Trametes rhubarb bidirectional fermentation system. The results showed:the growth of Trametes robiniophila Murr is in good condition, the mycoplasma dry discount rate and consumption rate in line with the requirements of the fermentation process, the mycoplasma polysaccharide content changes confirmed its existence to the material transformation, fermentation combination the Trametes robiniophila Murr and rhubarb constitute reasonable and feasible.(2)Rhubarb as fermentation substrate, bacteria growth, dry discount rate and consumption rate, mycoplasma polysaccharide content, content of anthraquinone as indicators examine the effect of amount of water, medicinal dosage, fermentation temperature to the bidirectional fermentation of Trametes rhubarb. Results determine the optimal fermentation process. The results showed that:the best fermented herbs plus water is120%, the best fermentation herbs dosage is10g (500mL conical flask),the best fermentation temperature is34℃.(3)Extracted anthraquinone from the Trametes rhubarb bidirectional fermentation system and the original medicinal rhubarb, through free anthraquinone, total anthraquinone and bound anthraquinone content changes, as well as thin-layer chromatography method to compare rhubarb pharmacodynamic material change, and cell experiments preliminarily investigate its pharmacodynamic improvement. The results showed:after fermentation, bound anthraquinone content decreased significantly, free type anthraquinone content increased significantly, the fermentation method enables rhubarb-bound anthraquinone to free type anthraquinone, thereby easing the rhubarb effect to diarrhea,and there are new ingredients generates after fermentation of Trametes rhubarb bidirectional fermentation system.The Trametes rhubarb bidirectional fermentation system has effect of inhibition of tumor.
