Objective:To study the muscle spindle histological and morphological changes in Tumbar vertebral instability patients, to discuss the role of the muscle spindle in the occurrence and development process of lumbar vertebral instability, and to provide the theory basis for prevention and rehabilitation of the instability of the lumbar spine.Methods:The normal tissues are from the body of specimen fixed by formaldehyde, sharp dissection and cut off the muscle tissue in the lumbar region;Pathology tissues are from the patients that got lumbar vertebral instability and had operations in lumbar spine surgery of Affiliated Hospital of Qingdao University. All tissues do conventional paraffin section, for HE dyeing and immunohistochemical to study the muscle spindle organization form, and count the immunohistochemical positive reflection.Results:Lumbar instability group shows pathological changes of structure in varying degrees, mainly for the increase in size, muscle spindle capsule enlargement and the cavity muscle spindle fibrous connective tissue capsule becoming thick; many muscle spindles existing clusterly; intrafusal muscle fiber reducing or replaced by granulation tissue.Between the two groups muscle spindle density has no significant difference (q=0.112and0.597, P>0.05);Under microscope to see the two groups of muscle spindle are NT-3positive expression, but the lumbar spine instability group shows low intensity dyeing, and there is a significant difference significance (q=3.507~23.663, P<0.01);The control group, the nuclear bag fiber was MY-32positive reflection, and lumbar spinal instability groups, nuclear bag fiber on MY-32dyeing intensity was decreased, the differences are significant.(q=3.509~23.673, P<0.01).Conclusions:Lumbar muscle spindle plays an important role in the origination and development of the lumbar instability. This effect did not reflect in quantity, but mainly reflect in the physiological function which can incarnate from the difference of the express of NT-3and MY-32and the change of morphology. The physiological function of muscle spindle depends on the normal of the structure and muscle fiber myosin heavy chain of phenotypic normal.NT-3is the necessary neurotrophy factor of the physiological function of muscle spindle. The change of morphological of the muscle spindle and the expression of NT-3, MY-32declares that muscle spindle plays an important role in the instability of lumbar instability.
