Background:The vulnerable plaques broking and secondary thrombosis were the main cause of acute cardiovascular incidents. Stabilizing the vulnerable plaques would help to reduce the acute cardiovascular incidents. Therefore, stabilizing the vulnerable plaques is becoming a hot issue in the field of cardiovascular disease prevention and therapy. We were also very interested in it and had proved that QingReQuYu granula (QRQYG) could stabilize vulnerable plaques in our previous study. But its functionary target point and its mechanism were needed to explore forward. So we choose to do the study of QRQYG stabilizing plaques through inhibiting the expression of Tenascin-C in patients, thus providing scientific basis and new ideas for traditional Chinese medicine in the treatment of stabilizing the vulnerable plaques.Objective:To investigate the function of QingReQuYu granula inhibiting the expression of CD3, CD68, ICAM-1, CD40L, MMP9, Tenascin-C on the carotid atherosclerotic plaque in the patients with severity carotid stenosis.Methods:We classified62patients with severity carotid stenosis into the treatment group(n=31)and control group(n=31) randomly.The patients in treatment group were given QingReQuYu granula and western medicine.The control group were given western medicine only.The course of treatment was16weeks. All of the patients were accepted endarterectomy.Plaques were subjected to analysis of CD3, CD68, ICAM-1, CD40L, MMP9, Tenascin-C, and collagen content by immunocytochemistry or polarized light analysis.Results:By the end of the experiment, the reduction of the expression of the CD3, CD68, ICAM-1, CD40L, MMP9, Tenascin-C in the plaques were statistic significant in the experiment group in comparing with that of the control group.Conclusions:The QingReQuYu granula can stabilize the carotid artery plaques through inhibiting the pro-inflammation factors and inhibiting the Tenascin-C, MMP9pathway.
