Muliple sclerosis (MS) is a T cell mediated chronic inflammatory demyelinating disorder of the central nervous system(CNS) which involves autoimmune response. MS occurs more freguently in young adults, by having the recurrent attacks repeatedly, the disease is gradually exacerbated and resulted to disability in most patient that suffering from this disease, and because the current clinical treatments are not still satisfied, thereby MS is still considered as an intractable disease. Hence finding safe and effective treatment of MS are the important issues for today’s medical profession. Experimental autoimmune encephalomyelitis(EAE) is similar to human MS. it shows resemblances with MS for clinical, immunological, biochemical and pathological features, so as ideal model of MS, it has been used for studying MS mechanisms. even more so than for finding the treatments of MS. Acupuncture treatment is characterized by its integrity and two-ways directivity, it makes the immune system to maintain a relatively balanced state, which may be important mechanism of acupuncture treatment of MS, but regardless of acupuncture treatment for MS clinical studies or EAE experimental studies have not yet been verified. Consequently, this experiment in the use of acupuncture treatment on the basis of the establishment of the EAE model to explore the therapeutic effect of acupuncture on EAE and their possible mechanisms, and hormone therapy for the negative control, then detecting the levels of Interleukin-17(IL-17) and Nitric Oxide(NO) in the rats serum to verify the scientific basis of the effectiveness of acupuncture treatment to MS.Methods:43female wistar rats were randomly divided into normal group, EAE model group, the hormone therapy group and the acupuncture therapy group. EAE induction was performed on day0in EAE model group, the hormone therapy group and the acupuncture therapy group. Each rat from these groups was immunized by subcutaneous injection of a500μl oil-water emulsion made by mixing the guinea pig spinal cord homogenate and complete Freund’s adjuvant(CFA), to improve morbidty also added by the intrapertoneal injection of Bordetella pertussis within72h. It also stimulated to given to normal group rats by injected0.9%NaCl solution. The normal group and the EAE model group without any treatment until after induction of modeling the30th day. To prove the effectiveness of acupuncture treatment, the treatment with hormone therapy and acupuncture therapy were compared as positive and negative control. Treatment of the rats began10days after the induction of EAE model until after induction of modeling the30th day, as negative control, the hormone therapy group was received at once daily30mg/kg dose methylprednisolone sodium succinate solution by intravenous injection for3days consecutively. After treatment for methylprednisolone, continually given to each rat by stomach perfusion3ml of prednisolone acetate suspension per day. The acupuncture therapy group as positive control was acupunctured by Baihui(DU20), bilateral Shenshu(BL23) and Zusanli(ST36). Since the10th day after the induction of EAE model, the rats were observed their condition daily, and daily recorded their weight change and neurobehavioral score that according to Kono and others determinded5point fraction method. Gathering materials as the result of this experiment was prepared in the30th day after the induction of EAE model, all serums gathered by centrifugation from rat blood that taken by the decapitation method and taken the supernatant, and measured IL-17levels in serum by ELISA, then using nitrate reduction method for determination of the content of NO in the serum.Result:Through the neurobehavioral score indicate its successful modeling. In the neurobehavioral score, both therapy groups that include the hormone therapy group and the acupuncture therapy group compared with the EAE model group, shows significant reduction, the symptoms improved significantly(P<0.05). After treatment, the levels of IL-17and NO in serum of both therapy groups decreased significantly compared to the EAE model group(P<0.05). However, the regardless of difference between the hormone therapy group and the acupuncture therapy group for the neurobehavior score or the levels of IL-17and NO in serum have no significant difference statistically(P>0.05).Conclusion:In the EAE model, the cytokine IL-17and NO is closely related with Thl/Th2and Th17immune responses. Through intervention that the acupuncture treatment and hormone therapy in EAE model, it was found IL-17and NO content in rat serum is decreased after treatment, and in remission from that results can suggest to make to maintain functional balances between Thl/Th2and Th17/Treg. to achieve therapeutic purpose that being immune factors in body maintained at a relatively normal range, these results are showing that acupuncture treatment of multiple sclerosis have a exact therapeutic effect. Effecacy between both therapy groups is no significant differences statistically, indicate that acupuncture treatment and hormonal treatment of EAE have the same efficacy, however, the examples obtained by the hormone therapy group were two cases of death in treatment, to illustrate the acupuncture treatment is more safer than hormone therapy and no have side effects.
