As one of the main active material of Hericium erinaceus, a large number of medicine and pharmacology study suggests that HEP can improve immunity, anti-tumor, anti-aging and anti-lipidemic. Drugs and health-care products made by HEP is very popular with good prospects for development. This dissertation focused on the study on the immune regulation effect of HEP and its primary mechanism.Objective:1. To study the effect of HEP on the non-specific immune functions, humoral immune functions and cellular immune functions of immuno-suppressed mice.2. To study the effect of HEP on the Splenic cell metabolic activity, distribution of T lymphocyte subsets and the secretion of cytokine of normal mice.Method:1. Established immunosuppression animal model by cyclophosphamide induction. Weighed the immunity organs to measured the thymus index and spleen index; used the clearance of charcoal particles method to measured mice’s mononuclear macrophages functions; used sheep red blood cells immunizations to measured mice’s humoral immune functions;observed the delayed type hypersensitivity (DTH) induced by DNCB to measured mice’s cellular immune functions.2. Vitro culture spleen cell of normal mice to measured the cytotoxicity of NK cell and the proliferative response of lymphocyte induce by Con A; used flow cytometry method to measured the distribution of CD3+、CD4+、CD8+cell subsets in peripheral blood of normal mice; used Enzyme-linked Immuno Sorbent Assay(ELISA) to measured the IL-2、IL-4、IL-6、TNF-α、IFN-y level in blood serum of normal mice.Result:1. HEP can significantly improved the thymus index and spleen index, improved the mononuclear macrophages functions, increased the hemolysin level of blood serum and increased the delayed type hypersensitivity (DTH) level of immuno-suppressed mice.2. HEP can significantly improved the cytotoxicity of NK cell and the proliferation and differentiation function of mice’s lymphocyte induce by Con A, increased the percentage of CD3+, CD4+、CD19+cell and the ratio of CD4+/CD8+of peripheral blood, and promoted the secretion of IL-2、TNF-α IL-6、IFN-y but inhibited the secretion of IL-4. Conclusion:HEP can significantly improved the non-specific immune functions, humoral immune functions and cellular immune functions of immuno-suppressed mice. It also significantly increased the metabolic activity,the secretion of IL-2IL-6、 TNF-α、INF-y and the percentage of CD3+, CD4+、CD19+cell and the ratio of CD4+/CD8+of peripheral blood. On the basis of the studies above, HEP showed strong effect on immune function through multi-levels, multi-targets and multi-dimensions.
