| Major depressive disorder is a mental disorder characterized by depressed mood,anhedonia, and reduced energy. Depression is a major contributor to the global burdenof disease and disability with a high incidence.The current study showed that several physiological systems involved in thepathogenesis of depression, but drug therapy for the treatment of depression were alsowholly dependent on the antidepressants targeted the monoamine neurotransmittersystems. However, according to recent reports, these antidepressants showed partialeffect in the treatment of patients with depression after all, and also indicated sideeffects that could not be ignored. Existing research suggested that Panax ginseng,valerian, and omega-3, not only showed antidepressant-like effects, but also showedsynergy with other antidepressants. Combination therapy cannot simply play theadvantage of their composition. The compound recipe can improve the degree ofcompliance of the drug and possible withdrawal effects.In our study, these three components were mixed up with a certain proportion ofthe portfolio as an antidepressant compound recipe, to achieve the purpose ofcombination therapy. Classic antidepressant evaluation model, the forced swim and tailsuspension model to evaluate the acute antidepressant effects of this compound recipe,the long-term treatment of compound recipe and the pharmacological mechanismsinvolved were investigated by chronic mild stress model of depression.Our results showed that the compound recipe in the classic acute antidepressantmodel showed a significant antidepressant effect. In acute tail suspension test andforced swimming test compound recipe significantly reduced the proportion ofmotionless behavior. As the reference drug Imipramine, we found that the compoundrecipe does not reduce the locomotor activity in mice as Imipramine. The antidepressanteffect of the compound recipe did not affect mice the locomotor activity activity. InCMS, the compound recipe could effectively release the CMS-induced depressive symptoms, and reverse the decreased5-HT concentration and the increased incorticosterone caused by the CMS. The compound recipe and Imipramine showed asimilar effect on its antidepressant effect, while the compound recipe did not induce ashort-term sedative effect compared with Imipramine. The compound recipe indicatedthe antidepressant effect with smaller side-effects. |
