| Gynostemma pentaphyllum (Thunb.) Makino, which belongs to Cucurbitaceae, Gynostemma BL.Herbs, is a kind of Perennial herb liane.All of it can be made into medicine, cold sweet in nature. It can clear heat and expell miasma, dispell phlegm, arrest cough,reduce blood fat, enhance immunizing ability etc. As the one of the higher active components, Gynostemma pentaphyllum polysaccharides have shown significant clinical applications in antioxidation, anti-tumor,enhancement immunity and so on.Selenium(Se) is one of the16kinds of essential micronutrients for human. The recent studys shown it plays a crucial role in antioxidant, heavy metal antagonism, prevention cancer and anti-cancer, immunity improvement and so on. In China, two-thirds of the region belongs to the environment where Se is not enough, but in human body it mainly comes from food, can’t self-synthesis.Accordingly, we find it is necessary to satify the needs of Se immediately. In nature, Se has two existing forms: inorganic Se and organic Se, organic Se is better absorber by the body than inoganic Se. At present, the development of Se-riched products, especially Se-enriched plant polysaccharides, have already caused more and more research interestings, which can provide the theory basis and the practical basis for the improvement of the absorption level of Se, not only in China but also all over the world.In this study, we mainly detected the Se content in Gynostemma pentaphyllum from10different provinces. And through the correlation of Se-Soil, Se-arsenic(As), Se-mercury(Hg), Se absorption and utilization of Gynostemma pentaphyllum have been detected, and Se effection to individuals have been analysed. Finally, We screened an Selenium-riched Gynostemma pentaphyllum then did some researchs on it’s biological activities. The main results are as followed:1. In test analysis, the Se contents of wild and cultivated Gynostemma pentaphyllum was detected from Ankang, and the rusults showed that the Se contents are significantly different as the various organs, this conclusion plays a theoretical guidance role for us to choose the harvest parts of Gynostemma pentaphyllum; To the contrast of the bigger strains in present cultivation area from the GAP plantation base, in the same growth environment and cultivation mode, the Tetraploid4’s Se content in September was more than1mg·kg-1, so this variety can be used as selenium-rich Gynostemma pentaphyllum and be worth researching.2. The Se contents of wild and cultivated Gynostemma pentaphyllum was detected from Shannxi, Hubei, Hunan, Anhui, Yunnan, Sichuan, Chongqing, Jiangxi, Guizhou, Henan provinces, compared with the Se content of its rhizospheric soil, we found that Se of any wild Gynostemma pentaphyllum mainly comes from the Se in soil. In the contrary, Se of any cultivated Gynostemma pentaphyllum didn’t have dependment on the Se in soil, this phenomenon may due to fertilizations, the influences of atmospheric settlement from environment and so on.3. We detected As and Hg contents of wild and cultivated Gynostemma pentaphyllum from10provinces, and analysised the antagonism from Se to As and Hg in Gynostemma pentaphyllum, but the rusults show that they didn’t have any correlation. This conclusion may due to many reasons:the antagonism from Se to heavy metal was affected by a lot of factors, we could not analyse it only by the contents, but also take the soil, the heavy metal ions settlement from environment and the heavy metal enrichment from plant itself under consideration. This part of the research remains to be further exploration.4. Antioxidative abilities of Gynostemma pentaphyllum polysaccharide(GMP) and Selenium-riched Gynostemma pentaphyllum polysaccharide(Se-GMP) were separately analyzed for DPPH-,-OH radical-scavenging activities and Fe3+reductive abilities. The results showed that the GMP and Se-GMP have strongly scavengeing activities, and certain Fe3+reductive ability, which increased with the increasing of its concentrations. The two free radical-scavenging activities of the Se-GMP are higher than the GMP, What’s more, when the concentration was2.4mg·ml-1, scavenging rate of Se-GMP for·OH radical reached91.8%, it’s very close to the Vc’s scavenging rate97%. All the conclusions showed that the Se-GMP had good potential in the removal of-OH radical.5. Anti-lead properties were exhibited by the the GMP and Se-GMP. The results showed that, after animal stomach injection50,100,200mg·kg-130d, GMP had significantly reduced the blood lead level of mice, but the liver lead level was also higher than the normal levels. Se-GMP had obviously reduced the content of blood lead and liver lead, and the anti-lead properties increased with the increasing of its concentrations. The anti-lead properties of Se-GMP was investigated according to the Standard of Health Food Test and Asses, it has the functions of promoting the lead discharge. |
