| Objective:To observe the impact of transfection of bone morphagenetic protein2(BMP-2) gene mediated by hydroxyapatite(HA) nanoparticles with chitosan(CS) surfacemodification on the cell toxicity reaction of HA-CS nanoparticles carrier to theosteoblastã€the expression of BMP-2gene,proliferation and differentiation of osteoblast.Methods: Recovery and culture osteoblast; The surface of HA nanoparticles wasmodified by CS;It was detected the cell toxicity reaction of HA-CS nanoparticles carrierto the osteoblast by MTT assay.The BMP-2gene were transfected into osteoblast mediatedby HA-CS nanoparticles,to evaluate the transfection efficiency. RT-PCR was used toidentify the expression of BMP-2;The proliferation of osteoblast and the activity ofalkaline phoaphatase(ALP) were measured.Results: MTT assay demonstrated that HA-CS nanoparticles suspension liquid hadno significant effects on the growth of osteoblast at the concentration below500μg/ml,andtransfection efficiency by modified HA better than not. RT-PCR demonstrated in theosteoblast,the BMP-2gene was steadily expressed and more stronger compare withcontrol group and empty plasmid group;Increase Osteoblast proliferation and enhance thebone ALP group activity after transfectedConclusion: HA-CS nanoparticles to the Osteoblast non-toxic role at a certainconcentration. And as a carrier,purpose gene BMP-2could be safely transfected intoosteoblast mediated by it and expressed steadily,at the same time, promoting theproliferation and differentiation of osteoblasts. |
