The Kidney Water Decoction On Mesangial Cell Fn, The Influence Of Collagen Type â…£ Expression

Purpose：Observation the expression of fibronectin (FN) and typeⅣ collagen that vascular Angiotensin Ⅱ (Ang Ⅱ) on rat Glomerularmesangial cells, and the effect that more kidney decoction on thatexpression.Method：1.Method of cultured Glomerular mesangial cells culturedin vitro,seeding density is105/cm2,using different concentrationsof Ang Ⅱ as colony-stimulating factor, application of ELISA methodfor the determination of24h,48h,72h,10%FBS group,10%RAT groupand5%FBS+5%RAT group of fibronectin (FN), type Ⅳ collagen content,to determine the time, Ang Ⅱ concentration and substrate role inglomerular mesangial cells at the end of class;2. Apply serumpharmacological method for preparation of different pharmacologicalserum concentrations of more kidney decoction of kidney in rats andnormal sera;3. Use of Ang Ⅱ as colony-stimulating factor,determination the content of fibronectin (FN), type Ⅳ collagen among different concentration of more kidney decoction of kidney group andnormal group by ELISA method.Results：1.Ang Ⅱ can significantly promote the expression offibronectin (FN) and type Ⅳ collagen in rat Glomerular mesangialcells, and Ang Ⅱ in10-7mmol/L concentrations, stimulating48h,10%RAT group express most apparent;2. the content of fibronectin(FN) and type Ⅳ collagen in more kidney decoction of high-dose andlow-dose group significantly lower than case group, with statisticalsignificance (P<0.05), more kidney decoction on fibronectin (FN) andtype Ⅳ collagen expression down.Conclusion：Ang Ⅱ promotes the expression of fibronectin (FN) andtype Ⅳ collagen on Glomerular mesangial cells.More kidney decoctioncan significantly inhibit the expression of fibronectin (FN) and typeⅣ collagen renal on Glomerular mesangial cells.
