Objective:To investigate the therapeutic effect of promoting blood qushimethod on alpha-naphthylisothiocyanare (ANIT) induced intrahepatic cholestasisin rats.Methods:Forty rats were induced by ANIT to prepare the intrahepaticcholestasis model,and were randomly divided into modle group, positive controlgroup of western medicine and Chinese traditional medicine treatment group,besides twenty rats in moldle group,the other with ten rats in each group,andchoose ten normal SD rats as normal control group.All groups were once a dayto fill the stomach with medicine,the modle group and normal control group weretreated0.9%sodium chloride injection By6ml/kg, western medicine group wasgiven Adenosine methionine by0.1g/kg/d, Chinese traditional medicine group wasgiven promoting blood qushi method by2ml/d.The delivery time from the day ofmodeling, killing ten rats in modle group after froty-eight hours,leaving theserum for preparation. The rest rats were sacrificed after continuous seven daysfor treatment.Full automatic biochemical analyzer tested indexes of liverfunction (TBIL、DBIL、TBA、ALT、AST、ALP)in each group,enzyme-linked immunosorbentkits to determine the lipoprotein X (LP-X) in serum,the polymerase chain reactiontested Na~+-H~+exchanger in the mRNA expression of the body,observed pathologicalchanges in liver tissues under the light microscope,and transmission electronmicroscopy to observe the ultrastructure of liver cells.Results:⑴At the time of intrahepatic cholestasis, the level of serum totalbilirubin (TBIL),direct bilirubin (DBIL) and bile acid (TBA) in SD rat weresignificantly increased (P <0.05). Differently with48hours after moldling,Thelevel of TBIL、DBIL、TBA were decreased at the end of experiment(P<0.05).Afterexperiment completely,compared with moldle group,the level of TBIL、DBIL、alanineaminotransferase (ALT) were reducing in western medicine group and chinese traditional medicine group (P<0.05), However,no significant difference inefficacy between the two groups(P>0.05).Compared with moldle group,The levelofTBA and alkaline phosphatase(ALP)were were no significant differences in westernmedicine group(P>0.05),the level of TBA and ALP were decreased in Chinesetraditional medicine treatment group (P<0.05).The activity of AST was nosignificant difference in each group (P>0.05).⑵Compared with moldle group,thecontent of lipoprotein-X (LP-X) was significantly lower (P <0.05) in westernmedicine group and chinese traditional medicine group, there was no significantdifference (P>0.05).⑶At the time of intrahepatic cholestasis, the expressionof Na~+-H~+exchanger mRNA was decreased obviously, the western medicine group andchinese traditional medicine group can increase its expression, compared withmodel group, highly significant difference (P <0.01),but was still significantlylower than the normal group(P<0.01).⑷The hepatic lobule structure is incomplete,unclear contours,scattered spotty necrosis; bile duct epithelial cell swelling,necrosis,and lumen spacious;inflammatory cell infiltration of the portal areain model group by light microscopy.The structure of hepatic lobule is morecomplete,and only the individual liver cells showed vacuolar degeneration inwestern medicine group by light microscopy. Lobular architecture is relativelycomplete, interlobular bile duct were generally normal, a small amount ofinflammatory cells infiltrating portal area, liver cell degeneration and theextent of necrosis was lighter in chinese traditional medicine treatment groupby light microscopy.⑸Mitochondria within the cytoplasm were swelling,part ofthe mitochondrial cristae,and showed vacuolar degeneration;smooth endoplasmicreticulum did not change significantly, glycogen is more, golgi apparatushypertrophy;nucleus was marginated and larger,the heterochromatin was increasedin nuclear; visible kupffer’s cells in hepatic sinusoid, irregular in shape,a large number of lysosomes and mitochondria within the cytoplasm;liver cellorganelles were partly dissolved;bile canaliculy have different degrees ofexpansion, the microvillus of bile canaliculy were reduced,the tight zone orthe layer of ectoplasm around the bile duct was widened in model group by electron microscopy.Cytoplasm was dissolved,nuclear condensation, mitochondrialcontraction, smooth endoplasmic reticulum dilated, rough endoplasmic reticulumin western medicine group by electron microscopy.Liver cell structure was almostnormal, lots of organelles and glycogen granules in chinese traditional medicinetreatment group by electron microscopy.Conclusion:Promoting blood qushi method can improve abnormal serumbiochemical indicators at the time of intrahepatic cholestasis, raised the levelof LP-X, increased expression of Na~+-H~+exchanger mRNA, significantly reduced theliver cell degeneration, necrosis and capillary bile duct dilatation, for ANITinduced intrahepatic cholestasis in rats have a significant therapeutic effect.
