| Mulberry leaves is dried leaves of Moraceae plant, traditional Chinese medicine recorded that Mulberry Leaves can treat diabetes.Recent pharmacology studies indicate that Polyhydroxy alkaloids1-deoxojirimycin(DNJ) is one of the effective components of Mulberry leaves, its structure is similar to natural sugar,competitive inhibition of a-glucosidase,has significant hypoglycemic effect, Therefore, Research about DNJ in mulberry leaves attractsed more attention by scholars at home and abroad..Dried Mulberry leaf powder as raw materials in this paper, researched on Mulberry leaves DNJ detection, extraction, isolation, purification, and hypoglycemic effects of Mulberry leaf extract, DNJ purification of hypoglycemic effect in vitro on levels, main content and conclusions of the study as follows:1. The TLC method was established analysize DNJ. Silica gel G plate, n-butanol:acetic acid:water (4.5:1.2:1.2) as a developing agent, chloro-o-toluidine for reagent.The results showed that DNJ can be separated from other compound, spots was round and clear, the mothed was simple and sensitive, reproducibility well.rapid qualitative and quantitative analysis of rough available for DNJ.A high performance liquid chromatographic(HPLC) method fort he determination of DNJ was established by fluorimetric detection after precolumn derivatization with9-fluorenylmethyl chlorformate(FMOC-Cl), and determined DNJ content different parts of a mulberry tree. The method showed good linear relationship in the range of0.1-10μg/mL,the correlation coefficient was0.9993.The results showed that the content of DNJ in leaves of Morus alba L.was0.223%.The reaction rate was quick,the operation was easy and accuracy,and the derivative was stable.The content of DNJ in mulberry barkã€mulberry branch xylemã€cortex moriã€mulberry and silkworm excrement was determined by HPLC. The results showed that the contents of DNJ in mulberry bark was highest,had0.388%, next mulberry, the contents of DNJ in mulberry branch xylem,only0.051%.2. Extraction DNJ from mulberry leaves in acid-water was optimized by single-factor test and orthogonal design, obtained the optimum extraction conditions:extraction time is2.5h, the extraction temperature is70℃, the liquid ratio of25:1mL/g,pH for2, under the conditions, extraction rate of DNJ was0.166%.Extraction condition of ultrasonic assisted extraction1-Deoxynojirimycin (DNJ) in m-ulberry leaf was optimized by the response surface method. The results showed that whe-n the extraction time at60min, extraction temperature at75℃, ultrasonic power at350W, liquid-materials ratio at16:1, extraction rate of DNJ could reach0.187%,the results line with the predict value of the model, this model can be effective forecasting and an-alysis extract DNJ from Mulberry leaves by ultrasound-assisted extraction.By comparing the acid-water extraction and ultrasonic-assisted extraction of extraction rate (0.166%ã€0.187%, respectively),obtained the ultrasonic assisted extraction of Mulberry leaves DNJ better than acid-water.3. This paper researched that Mulberry leaves extract of DNJ-rich on hypoglycemic effect to diabetic mice in vivo, results displayed that with experimental process,symptoms of "san duo a litte" of diabetes mice was remited gradually, and it was very clear of drop blood glucose effect, extraction dose more high, drop blood glucose effect more good, high dose drop blood glucose effect was the best, rendering the dose-effect relationship, and better than acarbose. This may be associated with the hypoglycemic effect of DNJ.4. Studied on purification of DNJ by cation-exchange resin(001X7,H+), and drawed penetration curves of DNJ. The results showed that DNJ can flowed out when pH>9through the resin. At this point, color of the outflow liquid burned. Therefore, pH and color of the outflow liquid can indicated DNJ through resin.Purification of Mulberry leaves DNJ by optimum conditions, DNJ purity reached3.32%, then purity up to8.24%by anion exchange resins, and up to15.18%after ultrafiltration,. The results purity of DNJ has been improved significantly.The samples that purity of15.18%treated heat, found that DNJ had good thermal stability. The results laied the theoretical basis for the development of DNJ-rich health food with the hypoglycemic effect of heat sterilization.5. This paper researched that in vitro and excised organs,the effect that DNJ purification inhibited α-glucosidase.The results indicates that in vitro and excised organs,DNJ has good inhibition effect on a-glucosidase in intestinal mucosa, and DNJ purity more high, inhibition more strong,when DNJ purity in15%around, its inhibition effect better than acarbose. |
